目的:观察肾衰灵对慢性肾衰(CRF)大鼠肾组织层粘连蛋白(LN)、Ⅲ型胶原(C-Ⅲ)及转化生长因子β1(TGF-β1)mRNA表达的影响。方法:采用5/6肾切除法建立慢性肾衰肾小球硬化大鼠模型,随机分为正常组、模型组、赖诺普利组、肾衰灵组,8周后测定各组大鼠肾组织LN,C-Ⅲ及TGF-β1 mRNA表达及肾脏病理变化。结果:模型组大鼠肾组织LN,C-Ⅲ及TGF-β1mRNA表达与正常组相比均明显增加(P<0.05),肾衰灵、赖诺普利组明显低于模型组(P<0.05),肾衰灵组明显低于赖诺普利组(P<0.05)。肾衰灵组肾脏病理变化明显减轻。结论:肾衰灵对5/6肾切除大鼠肾脏有明显保护作用,肾衰灵可显著降低大鼠肾组织TGF-β1相对mRNA表达,使LN,C-Ⅲ型胶原的合成减少,减轻肾组织中细胞外基质的沉积和肾小球的硬化,进而达到延缓肾功能衰竭的目的。
Objective: To observe the effects of Shenshuanyin on the expression of laminin (LN), collagen Ⅲ (C-Ⅲ) and transforming growth factor-β1 (TGF-β1) mRNA in the kidney of chronic renal failure (CRF) rats. Methods: A rat model of chronic renal failure glomerulosclerosis was established by 5/6 nephrectomy. The rats were randomly divided into normal group, model group, lisinopril group and Shenbuiling group. After 8 weeks, Tissues LN, C-Ⅲ and TGF-β1 mRNA expression and renal pathological changes. Results: The expression of LN, C-Ⅲ and TGF-β1 mRNA in renal tissue of rats in model group were significantly increased compared with those in normal group (P <0.05), and were significantly lower in model group than in model group (P <0.05) ), Shenshuayuan group was significantly lower than lisinopril group (P <0.05). Renal failure group renal pathological changes significantly reduced. CONCLUSION: Shenshuanyin can obviously protect the kidneys of 5/6 nephrectomized rats. Shenshenling can significantly reduce the mRNA expression of TGF-β1 in rat kidney and decrease the synthesis of LN and C-Ⅲ collagen, Tissue deposition of extracellular matrix and glomerular sclerosis, and thus achieve the purpose of retarding renal failure.
