目的探讨Smad7、组蛋白去乙酰化酶1(HDAC1)在小鼠马兜铃酸肾病(AAN)中的表达及两者在马兜铃酸肾病发生、发展中的作用。方法将32只体重(20±2)g的SPF(无菌)雄性昆明小鼠随机分为4组,其中3组为实验组,灌胃给予不同剂量的马兜铃酸(AA)Ⅰ,分别为4 mg/(kg.d)、8 mg/(kg.d)、16mg/(kg.d)。另设对照组,PBS灌胃。应用免疫组织化学、免疫印迹法及RT-PCR技术,检测AAN小鼠和正常对照组中Smad7和HDAC1的表达情况。结果 HDAC1阳性着色主要分布于细胞核上,且在AAN中的表达强度明显高于正常对照组。免疫印迹法检测和RT-PCR提示,Smad7在AAN中的表达低于对照组,而HDAC1的表达显著高于对照组。结论在AAN中Smad7低表达以及HDAC1高表达,提示Smad7和HDAC1可能在AAN的发生、发展中起重要作用。
Objective To investigate the expression of Smad7, histone deacetylase 1 (HDAC1) in aristolochic nephropathy (AAN) and the role of both in aristolochic acid nephropathy. Methods 32 SPF (sterile) male Kunming mice weighing 20 ± 2 g were randomly divided into 4 groups, of which 3 were experimental groups, and were given intragastric administration of different doses of aristolochic acid (AA) Ⅰ 4 mg / (kg.d), 8 mg / (kg.d), 16 mg / (kg.d). Another control group, PBS gavage. Immunohistochemistry, Western blotting and RT-PCR were used to detect the expression of Smad7 and HDAC1 in AAN mice and normal controls. Results The positive staining of HDAC1 mainly distributed in the nucleus, and the expression intensity in AAN was significantly higher than that in normal control group. Immunoblotting and RT-PCR suggested that the expression of Smad7 in AAN was lower than that in control group, while the expression of HDAC1 was significantly higher than that in control group. Conclusion The low expression of Smad7 and high expression of HDAC1 in AAN suggest that Smad7 and HDAC1 may play an important role in the development of AAN.
