目的研究原发性慢性闭角型青光眼(PCACG)患者小梁网组织中GRP78和CHOP的表达,探讨内质网应激在CACG小梁细胞氧化损伤中的作用。方法 HE染色法观察PCACG患者小梁组织的结构及形态变化。免疫组化检测PCACG患者小梁组织切片中的内质网应激相关因子GRP78、CHOP的表达水平。结果PCACG患者小梁网组织网眼窄小,部分伴有纤维化、消失,Schlemm管腔呈不规则性变窄,可见色素附着。免疫组化结果显示,GRP78、CHOP在对照组小梁组织中少量表达。而GRP78、CHOP在PCACG患者小梁组织中表达明显增加,差异均具有统计学意义(P<0.05)。结论 PCACG患者小梁网中存在慢性内质网应激,内质网应激可能参与了小梁细胞的氧化损伤。
Objective To investigate the expression of GRP78 and CHOP in trabecular meshwork cells of patients with primary chronic angle-closure glaucoma (PCACG) and to explore the role of endoplasmic reticulum stress in the oxidative damage of trabecular cells in CACG. Methods HE staining was used to observe the structure and morphological changes of trabecular tissue in PCACG patients. Immunohistochemistry was used to detect the expression of endoplasmic reticulum stress-related factors GRP78 and CHOP in trabecular tissue sections of PCACG patients. Results The mesh of trabecular meshwork was narrow in PCACG patients. Some of them were accompanied by fibrosis and disappeared. Schlemm’s lumen was narrowed irregularly. Pigment attachment was observed. The results of immunohistochemistry showed that GRP78 and CHOP were slightly expressed in the trabecular tissues of the control group. However, the expression of GRP78 and CHOP increased significantly in trabecular tissue of PCACG patients, with statistical significance (P <0.05). CONCLUSION: Chronic endoplasmic reticulum stress exists in the trabecular meshwork of patients with PCACG. Endoplasmic reticulum stress may be involved in the oxidative damage of trabecular meshwork cells.
