目的制备重组人干扰素-βSer17(rhIFN-βSer17),并检测其对SARS病毒的抑制作用。方法通过发酵收集菌体,经高压匀浆破碎收集包涵体、有机溶剂抽提、酸沉淀、柱层析纯化重组蛋白,通过细胞病变抑制法检测其对SARS病毒的抑制作用。结果研制的rhIFN-βSer17的纯度超过95%,比活性超过2.0×107IU/mg蛋白,在体外对SARS病毒具有明显的抑制作用。结论已成功制备rhIFN-βSer17,并在体外对SARS病毒具有抑制作用。
Objective To prepare recombinant human interferon-βSer17 (rhIFN-βSer17) and test its inhibitory effect on SARS virus. Methods The bacterial cells were harvested by fermentation. Inclusion bodies were harvested by high pressure homogenization, and the recombinant proteins were extracted by organic solvents, purified by acid precipitation and purified by column chromatography. The inhibitory effect on SARS virus was tested by cytopathic effect inhibition assay. Results The purity of rhIFN-βSer17 was over 95% and its specific activity was over 2.0 × 107IU / mg protein, which showed significant inhibitory effect on SARS virus in vitro. Conclusion rhIFN-βSer17 has been successfully prepared and has an inhibitory effect on SARS virus in vitro.
