肢体负压对动脉闭塞犬血浆内皮素和降钙素基因相关肽的影响

目的 观察肢体负压 (LNP)对肢体动脉闭塞犬血浆内皮素 (ET)和降钙素基因相关肽(CGRP)的影响 ,以探讨肢体负压的作用机理。方法 犬 15只 ,分治疗组 10只和对照组 5只 ,采用切断左后肢股动脉分支 ,动脉腔内置入螺旋状金属丝的方法 ,制作肢体动脉闭塞模型。 2周后 ,治疗组行患肢负压治疗 ,连续 10d ;对照组不做负压治疗。于模型制作前、制作后 2周及负压治疗 10d时 ,用放射免疫分析法检测患肢股静脉血浆ET及CGRP水平。结果 治疗组经负压治疗后 ,血浆ET水平显著低于治疗前 (P <0 0 1) ,血浆CGRP水平显著高于治疗前 (P <0 0 1) ,两者呈负相关 ;而对照组无明显变化 (P >0 0 5 )。结论 肢体负压可显著降低肢体动脉闭塞犬的血浆ET水平 ,提高CGRP水平。 Objective To observe the effects of limb negative pressure (LNP) on plasma endothelin (ET) and calcitonin gene related peptide (CGRP) in dogs with limb arterial occlusion to investigate the mechanism of limb negative pressure. Methods Fifteen dogs were randomly divided into treatment group (n = 10) and control group (n = 5). Artery occlusion model was made by cutting off branches of femoral artery of the left hind limb and inserting helical wire into the arterial cavity. After 2 weeks, the treatment group underwent negative pressure treatment for 10 days; the control group did not make negative pressure treatment. Before model making, 2 weeks after making and 10 days after negative pressure treatment, the levels of plasma ET and CGRP in the femoral vein were measured by radioimmunoassay. Results After treatment with negative pressure, the plasma ET level was significantly lower in the treatment group than before treatment (P <0.01), and the plasma CGRP level was significantly higher than that before treatment (P <0.01), while the control group No significant change (P> 0.05). Conclusion Limb negative pressure can significantly reduce the plasma ET level and improve the level of CGRP in dogs with arterial occlusive disease.
