淮安地区流感病毒流行特征及分子进化研究

目的了解淮安地区流感病毒流行及变异特点。方法选择一个流感监测年度(2014-2015年)分离的流感病毒,进行HA1基因的扩增、测序,采用PHYLIP 3.6软件及popart1.7软件构建系统进化树及Network进化网络图,利用DnaSP5.1软件进行病毒多态性分析,利用MEGA 6.0软件进行氨基酸推导及相关特征分析。结果 2014-2015年淮安地区流行的流感病毒核苷酸同源性及多样度分别为98.2%~99.9%和0.009(新甲型H1N1),97.3%~99.9%和0.010(H3N2),98.7%~99.8%和0.009(B型Yamagata系),其中B型流感Yamgata系为淮安地区优势株。新甲型H1N1流感病毒淮安株在进化树上形成一个独立的进化亚枝,由A/Beijing-Xingcheng-Nan-Pian/SWL11619分化形成;H3N2流感病毒淮安株形成3个进化亚枝,由A/HK/7409/2014辐射进化形成;B型Yamgata系流感病毒淮安株形成2个进化亚枝,由B/Zhejiang-Liandou/11000/2014或B/Jiangsu-Hailing/11443/2014分化形成。与疫苗株相比,淮安地区各型流感病毒均发生的变异:K180Q、S220T(新甲型H1N1),N294K(H3N2),H137Q、N144K(B型Yamagata系)。结论淮安地区一个流行年度有两次流感病毒流行高峰,与南方流行趋势一致。 Objective To understand the prevalence and variation of influenza virus in Huaian area. Methods The influenza virus isolated from the year of influenza surveillance (2014-2015) was selected for HA1 gene amplification and sequencing. Phylogenetic tree and network evolution graph were constructed using PHYLIP 3.6 software and popart1.7 software. Using DnaSP5.1 software Virus polymorphism analysis, the use of MEGA 6.0 software for amino acid deduction and related characteristics analysis. Results The nucleotide sequence homology and diversity of influenza viruses in Huai’an between 2014 and 2015 were 98.2% -99.9% and 0.009 (new H1N1), 97.3% -99.9% and 0.010 (H3N2) respectively, 98.7% 99.8% and 0.009 (Yamagata type B), of which Yamgata type B was the dominant strain in Huai’an. The new H1N1 influenza virus Huai’an strain formed an independent evolutionary sub branch in the evolutionary tree and was formed by the differentiation of A / Beijing-Xingcheng-Nan-Pian / SWL11619. The H3N2 influenza virus Huai’an strain formed three evolutionarily sub-branches, HK / 7409/2014 Radiation evolution formation; B-type Yamgata influenza virus Huai’an strain formed two evolution sub-branches, formed by B / Zhejiang-Liandou / 11000/2014 or B / Jiangsu-Hailing / 11443/2014. Compared with the vaccine strain, all kinds of influenza viruses in Huaian area were mutated: K180Q, S220T (new type A H1N1), N294K (H3N2), H137Q, and N144K (type B Yamagata). Conclusions There are two epidemic peak of influenza virus in a popular year in Huaian area, consistent with the trend in the south.
