【目的】利用正交实验优化桃儿七中鬼臼毒素的回流提取工艺。【方法】通过高效液相色谱法,Cosmosil C_(18)柱(2.5 mm,4.6 mm×250 mm),流动相甲醇水溶液梯度洗脱,292 nm下进行检测,建立鬼臼毒素的含量测定方法,以桃儿七中鬼臼毒素的含量及得率为评价指标。在单因素实验的基础上采用正交设计方法L9(34)考察提取温度、提取时间和料液比对鬼臼毒素提取效果的影响。【结果】桃儿七中鬼臼毒素的最佳提取工艺条件为:提取溶剂80%乙醇、提取2次、提取温度85℃、提取时间2.0 h、料液比1:10。通过验证试验,鬼臼毒素的总提取率可达86.62%,含量可达9.57%。【结论】经过优化后鬼臼毒素的提取工艺具有提取率高、工艺稳定、低成本等特点,该工艺条件可为其工业生产提供参考依据。
【Objective】 The orthogonal experiment was used to optimize the reflux extraction process of podophyllotoxin in peach seven. 【Method】 The method for the determination of podophyllotoxin was established by high performance liquid chromatography with Cosmosil C_ (18) column (2.5 mm, 4.6 mm × 250 mm) and gradient elution with mobile phase aqueous methanol solution at 292 nm. The content and yield of podophyllotoxin in peach seven were used as evaluation indexes. Based on the single factor experiment, orthogonal design method L9 (34) was used to investigate the effects of extraction temperature, extraction time and solid-liquid ratio on the extraction of podophyllotoxin. 【Result】 The optimum extraction conditions of podophyllotoxin in peach seven were as follows: extraction solvent 80% ethanol, extraction twice, extraction temperature 85 ℃, extraction time 2.0 h, solid-liquid ratio 1:10. Through the validation test, the total extraction rate of podophyllotoxin up to 86.62%, content up to 9.57%. 【Conclusion】 The optimized extraction process of podophyllotoxin has the characteristics of high extraction rate, stable process and low cost. The process conditions can provide a reference for its industrial production.
