目的:建立中药潜在护肝活性物质快速筛选新方法,辨识赤芍内具有护肝活性的化学成分。方法:在半乳糖胺HepG2细胞损伤模型上,采用二乙酸荧光素荧光标记法和MTT法筛选赤芍中具有护肝活性的组分,使用色谱-质谱联用技术分析其化学组成,并对筛选出的赤芍护肝活性成分进行实验验证。结果:从赤芍中筛选发现具有明显护肝活性的化学组分3个,并分析鉴定出芍药苷、棕榈酸乙酯和亚油酸乙酯3个成分。验证实验表明,这3个化合物对HepG2细胞均具有较明显的保护作用。结论:赤芍所含的芍药苷、棕榈酸乙酯和亚油酸乙酯具有潜在的护肝活性。
OBJECTIVE: To establish a new method for rapid screening of potential hepatoprotective active ingredients of traditional Chinese medicine (TCM) and to identify the chemical constituents with active hepatoprotective activity in Radix Paeoniae Rubra. METHODS: HepG2 cell injury model was established by galactosamine HepG2 cells. The components of Hepatitis C virus (Paeoniae Rubra) with hepatoprotective activity were screened by fluorescein diacetate staining and MTT assay. The chemical constituents were analyzed by GC-MS. Out of red peony Liver active ingredients for experimental verification. Results: Three chemical constituents with obvious hepatoprotective activities were screened from Radix Paeoniae Rubra and identified as paeoniflorin, ethyl palmitate and ethyl linoleate. Validation experiments show that these three compounds have a more significant protective effect on HepG2 cells. Conclusion: Paeoniflorin, ethyl palmitate and ethyl linoleate contained in Radix Paeoniae Alba have potential hepatoprotective activity.
