目的 探讨人β干扰素(IFN β)与胶质瘤细胞凋亡的关系及意义。方法 利用脂质体转染方法将IFN β真核表达载体pSV2IFN β导入SHG44胶质瘤细胞。细胞免疫荧光和免疫组化检测IFN β基因的稳定转染及表达,利用流式细胞仪、透射电镜观察细胞凋亡情况。结果 IFN β基因成功转染SHG44胶质瘤细胞并得以表达。转染细胞的凋亡细胞数与未转染细胞相比有显著性差异。结论 IFN β能够诱导人SHG44胶质瘤细胞凋亡。
Objective To investigate the relationship between human β interferon (IFN β) and apoptosis of glioma cells and its significance. Methods The eukaryotic expression vector pSV2IFNβ was transfected into SHG44 glioma cells by lipofection. Immunofluorescence and immunohistochemistry were used to detect the stable transfection and expression of IFN-β gene. Cell apoptosis was observed by flow cytometry and transmission electron microscopy. Results The IFNβ gene was successfully transfected into SHG44 glioma cells and expressed. The number of apoptotic cells in transfected cells was significantly different from that in untransfected cells. Conclusion IFNβ can induce human SHG44 glioma cell apoptosis.
