目的 从虾蛄壳提取壳聚糖 ,将α-淀粉酶固定在壳聚糖上。 方法 以自制壳聚糖为载体 ,一氯乙酸或三氯乙酸为交联剂 ,将α-淀粉酶固定化。 结果 三氯乙酸是较好的交联剂。固定化酶与原酶的最适 p H及最适温度基本相同 ,均为 6.5 ,5 0℃ ;固定化酶在 65℃仍有较高热稳定性 ,而原酶活力明显下降 ;固定化酶的表观米氏常数 Km =0 .6× 10 -2 g/ml,原酶 Km =1.1× 10 -2 g/ml;一定浓度 Cl-、Br-离子有助提高固定化酶活力。 结论 该固定化酶活力较高 ,热稳定性和贮存稳定性好 ,是良好的检测试剂。
Objective To extract chitosan from shrimp shell and fix the α-amylase on chitosan. Methods Chitosan as carrier, monochloroacetic acid or trichloroacetic acid as crosslinking agent, the α-amylase was immobilized. Trichloroacetic acid is a good cross-linking agent. The optimal pH and optimum temperature of the immobilized enzyme and the original enzyme were about 6.5 and 50 ℃ respectively. The immobilized enzyme still had higher thermal stability at 65 ℃, while the activity of the original enzyme decreased obviously. Apparent Michaelis constant Km = 0.6 × 10 -2 g / ml, the original enzyme Km = 1.1 × 10 -2 g / ml; a certain concentration of Cl-, Br- ions help to increase the activity of immobilized enzyme. Conclusion The immobilized enzyme has high activity, good thermal stability and storage stability, and is a good detection reagent.
