目的观察隔药饼灸对功能性胃肠病(FGIDs,Functional Gastrointestinal Disorders)肝郁脾虚型模型大鼠组织中5-HT_7及转运体(SERT,Serotonin Transporter)的影响,探讨隔药饼灸治疗FGIDs的可能作用机制。方法 48只SD大鼠按照随机数字表法随机分为空白组、模型组、隔药饼灸组、逍遥散组4组,每组12只,除空白组外,其余3组均采用复合病因法(慢性束缚应激+过度疲劳+饮食失节+夹尾+摇晃)连续造模21d,分组治疗2周后,用real time PCR法检测结肠组织中5-HT_7受体及转运体的表达。结果与模型组相比,隔药饼灸组和逍遥散组大鼠结肠组织中5-HT_7受体表达下降(P<0.05);SERT的表达明显升高(P<0.01),差异有统计学意义,但两组间比较差异无统计学意义(P>0.05)。结论隔药饼灸能和逍遥散一样通过下调结肠组织中5-HT_7受体的表达,以及上调结肠组织中转运体的表达,从而达到改善胃肠动力和感觉异常的作用,这可能是其治疗FGIDs的作用机制之一。
Objective To observe the effect of drug-cake-separated moxibustion on the expression of 5-HT_7 and transporter (SERT, Serotonin Transporter) in the rats with functional deficiency of the liver and spleen due to functional gastrointestinal disease (FGIDs) The possible mechanism of action. Methods 48 SD rats were randomly divided into blank group, model group, drug-cake moxibustion group and Xiaoyao powder group, with 12 rats in each group. Except for the blank group, the other three groups were treated by compound etiology (Chronic restraint stress + over-fatigue + dietary nodules + nip + shaking) for 21 days. After treatment for 2 weeks, the expression of 5-HT_7 receptor and transporter in colon tissue were detected by real time PCR. Results Compared with the model group, the expression of 5-HT 7 receptor in colon mucosa of moxibustion group and Xiaoyao powder group decreased (P <0.05), and the expression of SERT increased significantly (P <0.01), the difference was statistically significant However, there was no significant difference between the two groups (P> 0.05). Conclusions Cake-separated moxibustion can, like Xiaoyao San, improve gastrointestinal motility and sensory abnormalities by down-regulating the expression of 5-HT 7 receptor in colon tissues and up-regulating the expression of transporters in colon tissues, which may be the result of its treatment One of the mechanisms of action of FGIDs.
