目的通过血管紧张素Ⅱ(AngⅡ)刺激肝癌细胞系HEPG2,探讨其是否可以促进HEPG2发生上皮间质转化(EMT)。方法使用AngⅡ刺激HEPG2,进而采用蛋白印记法对HEPG2中Vimentin和E-cadherin蛋白表达水平进行检测;探讨其发生EMT的合适时间;并使用AngⅡ的抑制剂Ang1-7和Ang1-7的抑制剂A779进行干预,明确AngⅡ诱导HEPG2发生EMT的作用。结果 AngⅡ刺激下,HEPG2中E-cadherin转录水平下降,Vimentin转录水平升高;最适宜刺激时间是48 h。AngⅡ诱导HEPG2发生EMT的作用可部分被抑制剂Ang1-7抑制。结论 AngⅡ刺激下,HEPG2可以发生EMT。
Objective To investigate whether HEPG2 can induce epithelial-to-mesenchymal transition (EMT) in HEPG2 by stimulating hepatocellular carcinoma cell line HEPG2 with angiotensin Ⅱ (AngⅡ). Methods AngⅡ was used to stimulate HEPG2, and Western blotting was used to detect the expression of Vimentin and E-cadherin protein in HEPG2. The suitable time for EMT was also explored. AngⅡ inhibitor Ang7 and Ang7 inhibitor A779 Intervention, clear Ang Ⅱ induced HEPG2 the role of EMT. Results Under Ang Ⅱ stimulation, the transcription level of E-cadherin and the expression of Vimentin in HEPG2 increased, and the optimal stimulation time was 48 h. AngⅡ induced HEPG2 the role of EMT can be partially inhibited by inhibitor Ang1-7. Conclusion AngⅡ can induce EMT in HEPG2.
