目的:通过轮状病毒SA11株感染乳鼠为实验模型,研究七味白术散对肠道s Ig A、血清IFN-γ及小肠黏膜病理改变的影响,探讨其对小肠黏膜的保护作用。方法:给予灌胃病毒液的模型乳鼠连续灌胃七味白术散、病毒唑及生理盐水。采集血清及小肠标本,采用酶联免疫法(ELISA)检测乳鼠肠道s Ig A、血清IFN-γ的含量,对乳鼠空肠段做石蜡切片HE染色观察病理改变。结果:七味白术散组和病毒唑组乳鼠腹泻天数较少,与模型组比较,差异均有统计学意义(P<0.05)。七味白术散组和病毒唑组病毒清除率均为100%,与模型组比较,差异均有统计学意义(P<0.05)。七味白术散组和病毒唑组小肠黏液s Ig A较高,与模型组比较,差异均有统计学意义(P<0.05)。七味白术散组和病毒唑组IFN-γ升高,与模型组比较,差异均有统计学意义(P<0.05)。正常组小肠黏膜未见病理改变。模型组黏膜下层可见明显的炎性细胞浸润,黏膜上皮细胞有少量坏死脱落,肌层可见明显的肿胀。病毒唑组可见黏膜下层结构疏松,稍许水肿,黏膜层上皮细胞未发现明显病理改变;肌层间些许肿胀,可见空染的区域和炎性细胞浸润。七味白术散组小肠黏膜结构正常,未见明显病理改变。结论:七味白术散能加快对病毒的清除,改善腹泻症状,减轻对小肠黏膜的损害。
OBJECTIVE: To investigate the effect of Qiweibaizhu San on pathological changes of intestinal s Ig A, serum IFN-γ and small intestinal mucosa in mice infected by rotavirus strain SA11 and explore its protective effect on intestinal mucosa. Methods: The model rats given intragastric inoculation of virus solution were continuously fed with Qiweibaizhu San, Ribavirin and normal saline. Serum and small intestine specimens were collected. The contents of s Ig A and IFN-γ in the intestine were detected by enzyme-linked immunosorbent assay (ELISA). The pathological changes were observed by paraffin sections HE staining. Results: The number of diarrhea days was less in the rats treated with Qiweibaizhu Powder and Ribavirin, compared with the model group, the difference was statistically significant (P <0.05). The clearance rate of virus in both the Baishasai Baizhu San and Ribavirin groups was 100%, which was significantly different from the model group (P <0.05). Compared with model group, the differences of s Ig A in intestinal mucosa between Qiweibaizhufa group and ribavirin group were statistically significant (P <0.05). Compared with model group, the differences of IFN-γin Qiwei Baizhu San and Ribavirin groups were statistically significant (P <0.05). The normal group of small intestinal mucosa no pathological changes. The model group mucosal visible inflammatory cell infiltration, mucosal epithelial cells with a small amount of necrosis, muscular visible swelling. Ribavirin group showed submucosal structure loose, a little edema, mucosal epithelial cells found no significant pathological changes; a slight swelling between the muscular layer, visible empty areas and inflammatory cell infiltration. Qiwei Baizhu San group normal intestinal mucosa structure, no significant pathological changes. Conclusion: Qiweibaizhu Powder can speed up the removal of the virus to improve the symptoms of diarrhea, reduce damage to the small intestine mucosa.
