目的阐明心血管组织是否存在局部肾素 -血管紧张素系统 (localrenin angiotensinsystem ,L RAS)主要成份的表达。方法提取大鼠左室心肌、基底动脉、颈总动脉、腹主动脉和股动脉组织总RNA ,以G3PDH基因表达作为内参照 ,用逆转录酶 -多聚酶链式反应 (reversetranscriptase polymerasechainreaction ,RT PCR)方法 ,半定量检测上述组织血管紧张素原 (angiotensinogen ,AGT)和血管紧张素转化酶 (angiotensin convertingenzyme ,ACE)基因表达水平。 结果大鼠左室心肌、基底动脉、颈总动脉、腹主动脉和股动脉组织均有AGT和ACEmRNA表达。ACEmRNA在基底动脉和腹主动脉组织的表达水平显著高于颈总动脉和股动脉 ,而不同血管间AGTmRNA的表达水平则无显著差别。结论生理条件下大鼠心肌和不同部位大、中动脉血管组织均存在局部RAS主要成份AGT和ACE基因的表达 ,支持心血管组织存在L RAS这一重要观点
Objective To elucidate the expression of major components of local renin - angiotensin system (L RAS) in cardiovascular tissue. Methods Total RNA was extracted from the left ventricular myocardium, basilar artery, common carotid artery, abdominal aorta and femoral artery of rats. G3PDH gene expression was used as an internal control, and reverse transcriptase polymerase chain reaction (RT PCR) The mRNA levels of angiotensinogen (AGT) and angiotensin-converting enzyme (ACE) were detected by semi-quantitative analysis. Results The AGT and ACE mRNA expressions in the left ventricular myocardium, basilar artery, common carotid artery, abdominal aorta and femoral artery tissue of rats. ACE mRNA expression in basilar artery and abdominal aorta tissue was significantly higher than that in common carotid artery and femoral artery, while there was no significant difference in the expression level of AGT mRNA between different blood vessels. Conclusion Physiological conditions of rat and large and middle cerebral artery in different parts of the existence of the main components of local RAS AGT and ACE gene expression, support the existence of cardiovascular R LAS this important point
