目的考察湖北贝母中生物碱在蒸发光散射检测器(ELSD)中的响应因子是否一致,进而利用ELSD响应因子的一致性建立湖北贝母的含量测定方法,同时测定湖贝甲素、贝母素乙和贝母素甲的含量。方法采用Thermo Acclaim 120C18(4.6mm×250mm,5μm)色谱柱,流动相:乙腈-0.02%二乙胺溶液(74∶26),流速1.0 ml·min-1,检测器为Alltech2000ES ELSD,漂移管温度为93℃,载气流速为2.4L·min-1,对数外标两点法定量。结果通过测定3种生物碱的线性方程,考察其响应因子。3种物质的线性方程间无显著性差异(P>0.05)。结论湖北贝母中生物碱成分在ELSD中的响应因子一致,可尝试用“一测多评”的质量评价模式用于湖北贝母药材的质量控制。
Objective To investigate whether the response factors of alkaloids from Fritillaria cirrhosa L. in Evaporative Light Scattering Detector (ELSD) are the same, and to establish the method of determination of Fritillaria cirrhosa by ELSD response factor, Vegetarian B and peimine A content. Methods A Thermo Acclaim 120C18 (4.6 mm × 250 mm, 5 μm) column was used. The mobile phase consisted of acetonitrile and 0.02% diethylamine (74:26), the flow rate was 1.0 ml · min -1 and the detector was Alltech 2000 ES ELSD. 93 ℃, the carrier gas flow rate of 2.4L · min-1, the number of external standard two-point method. Results By measuring the linear equations of three alkaloids, the response factors were investigated. There was no significant difference between the three equations (P> 0.05). Conclusion The response coefficients of the alkaloids in Fritillaria hirsutum L. in ELSD are the same, and the quality evaluation model of “one test for multiple evaluation” can be used for the quality control of Hubei Fritillaria.
