目的研究乐果对人外周血淋巴细胞热应激蛋白70(HSP70)表达的影响,初步探讨HSP70用于有机磷农药生产的环境及生物监测的可能性。方法选取健康对照者11人及不同工种的乐果接触工人35人,分离其外周血淋巴细胞后,用流式细胞仪检测HSP70基础表达及经乐果体外诱导后水平,同时检测其全血乙酰胆碱酯酶(AChE)活力,分析其可能的影响因素。结果接触者与对照者外周血淋巴细胞HSP70基础表达水平分别为41.24%±10.45%和23.97%±4.29%,AChE活力分别为(125.23±7.97)、(145.36±8.78)U/ml,差异均有统计学意义(P<0.01)。不同工种乐果接触工人其外周血淋巴细胞HSP70基础表达水平分别为47.34%±11.87%和38.05%±8.20%,差异有统计学意义(P<0.05),但AChE活力差异无统计学意义(P>0.05)。对接触者HSP70基础表达水平影响有统计学意义的因素按程度大小依次为:健康状况、环境浓度和工龄,它们共可解释HSP70至少88%的变异;对AChE活力影响有统计学意义的因素只有工龄,其只能解释AChE约12%的变异。经体外乐果诱导后,对照组HSP70诱导水平较接触组高,差异有统计学意义(P<0.01)。不同人群淋巴细胞HSP70表达增加幅度不同,按其大小依次为:对照组、操作工和包装工。对HSP70变化率影响有统计学意义的因素为环境浓度和工龄。结论HSP70可作为综合反映乐果接触工人自身状况及所处作业环境的指标。
Objective To study the effect of dimethoate on the expression of heat shock protein 70 (HSP70) in human peripheral blood lymphocytes and to explore the possibility of environmental and biological monitoring of HSP70 for organophosphorus pesticide production. Methods 11 healthy controls and 35 workers with dimethoate in different kinds of work were selected. After the peripheral blood lymphocytes were isolated, the basal expression of HSP70 and the induced level of HSP70 in vitro were detected by flow cytometry. Meanwhile, the levels of acetylcholine Esterase (AChE) activity, analysis of its possible influencing factors. Results The basal expression levels of HSP70 in peripheral blood lymphocytes in contact and control subjects were 41.24% ± 10.45% and 23.97% ± 4.29%, respectively. The AChE activities were (125.23 ± 7.97) and (145.36 ± 8.78) U / ml, the difference was statistically significant (P <0.01). The basal expression of HSP70 in peripheral blood lymphocytes of workers exposed to dimethoate was 47.34% ± 11.87% and 38.05% ± 8.20%, respectively, with statistical significance (P <0.05) However, there was no significant difference in AChE activity (P> 0.05). The factors influencing the basic expression level of HSP70 in contact persons were, in order of degree, health status, environmental concentration and length of service, which explained a total of at least 88% variation of HSP70. The factors influencing AChE activity were statistically significant Length of service, which can only explain about 12% of AChE variation. Induction of HSP70 in the control group was higher than that in the exposure group (P <0.01). Different groups of lymphocyte HSP70 expression increased in different degrees, according to their size in order: the control group, operators and packers. The factors influencing the rate of change of HSP70 with statistical significance were environmental concentration and length of service. Conclusion HSP70 can be used as a comprehensive indicator of the status of workers exposed to dimethoate and their work environment indicators.
