目的:探讨增液布津汤对干燥综合征(SS)小鼠细胞因子γ干扰素(IFN-γ)、白细胞介素6(IL-6)、白细胞介素1(IL-1)的影响,探寻中药对SS干预作用机理。方法:取70只BALB/c小鼠,按体重随机分为正常组、模型组、六味地黄丸组、增液布津汤小剂量组、增液布津汤中剂量组、增液布津汤大剂量组共6组,各给药组均在第0天即开始给药,每天1次,直至第60天。于首次免疫后第61天球后静脉放血处死小鼠,解剖取相应脏器及检测外周血细胞因子。结果:与正常组比较,模型组IL-1、IFN-γ、IL-6均显著增加(P<0.01)。而给予药物后,增液布津汤各剂量组的IL-1、IFN-γ、IL-6含量下降,与模型组比较,差异有显著性或非常显著性意义(P<0.05,P<0.01)。增液布津汤小剂量组IFN-γ/IL-6较模型组下降,差异有显著性意义(P<0.05)。结论:增液布津汤通过抑制细胞因子IL-6、IFN-γ蛋白合成及分泌,间接地抑制了Th细胞的分化与增殖,调节Th1/Th2平衡来治疗SS,从而缓解SS小鼠颌下腺淋巴细胞浸润,进一步说明细胞因子在SS唾液分泌过程中发挥重要作用。
Objective: To investigate the effects of Zeng Bu Bu Jin Tang on cytokine IFN-γ, IL-6 and IL-1 in mice with Sjogren’s Syndrome (SS) Mechanism of Chinese medicine on SS intervention. Methods: Seventy BALB / c mice were randomly divided into normal group, model group, Liuweidihuangwan group, low dosage Zengbu Bujin decoction, middle dosage Zengbu Bu decoction, high dosage Zengbu Bu decoction A total of 6 groups, each treatment group were started on the 0th day of administration, once a day, until the first 60 days. On the 61st day after the first immunization, the mice were sacrificed by exsanguination of the posterior venules and the corresponding organs were dissected and the peripheral blood cytokines were detected. Results: Compared with the normal group, the levels of IL-1, IFN-γ and IL-6 in model group were significantly increased (P <0.01). Compared with the model group, the levels of IL-1, IFN-γ and IL-6 in the Zengbu Bujin decoction group were significantly decreased (P <0.05, P <0.01) . Compared with the model group, the concentration of IFN-γ / IL-6 in the Zengbu Bu Jin Tang low dose group was significantly lower (P <0.05). Conclusion: Zeng Bu Bu Jin Tang can inhibit the differentiation and proliferation of Th cells indirectly by inhibiting the synthesis and secretion of cytokines IL-6 and IFN-γ, and regulate Th1 / Th2 balance to treat SS, thus relieving the submandibular gland lymphocytes Infiltration, further instructions cytokines play an important role in the secretion of SS saliva.
