目的利用HCV多表位复合抗原免疫家兔制备高效价抗HCV多克隆抗体,为改进丙型肝炎病毒抗原的诊断试剂盒提供必要条件。方法HCV多表位复合抗原与完全弗氏佐剂混合免疫家兔,获得高效价抗血清,经硫酸铵沉淀及PrintA/G亲和层析纯化。结果已获得效价大于1∶16000的抗HCV多克隆抗血清,经纯化后达电泳纯。该抗体能与HCVNS3、NS5及C抗原特异性结合。结论HCV多表位复合抗原具有较好的免疫原性,可用于改进HCV病毒抗原诊断试剂。
Objective To prepare high-titer anti-HCV polyclonal antibody from rabbits by using multi-epitope antigens of HCV to provide the necessary conditions for improving the diagnostic kit of hepatitis C virus antigens. Methods Rabbit was immunized with HCV multi-epitope antigen and complete Freund’s adjuvant to obtain high-titer antiserum, and purified by ammonium sulfate precipitation and PrintA / G affinity chromatography. Results An anti-HCV polyclonal antiserum with titer greater than 1:16000 was obtained and purified to electrophoretic purity. The antibody specifically binds to HCV NS3, NS5 and C antigens. Conclusion The HCV multi-epitope antigen has good immunogenicity and can be used to improve HCV viral antigen diagnostic reagents.
