目的 描绘鼻咽癌细胞株不同转移潜能亚株的基因表达谱 ,以筛选鼻咽癌新的肿瘤相关及转移相关候选基因。方法 用含 14 0 0 0个cDNA克隆的表达微阵列分析鼻咽癌细胞株SUNE 1的 3个亚株 ,即高转移潜能细胞株 5 8F、成瘤不转移株 6 10B和不成瘤株 13 9B的mRNA表达 ;用半定量RT PCR验证cDNA表达微阵列检测的结果。结果 5 8F与 13 9B比较 ,有 82个差异表达基因 ;6 10B与 13 9B比较 ,有 38个差异表达基因 ;5 8F与 6 10B比较 ,有 5 4个差异表达基因 ;5 8F及 6 10B与13 9B比较 ,有 12个共有的差异表达基因 ;5 8F与 13 9B及 6 10B比较 ,有 14个共有的差异表达基因。这些基因涉及代谢、转录、分化、凋亡和信号转导等多种生物功能以及许多未知功能。结论揭示了鼻咽癌细胞株的基因表达谱 ,为寻找鼻咽癌相关基因提供了重要线索
Objective To characterize the gene expression profiles of different metastatic potential subtypes of nasopharyngeal carcinoma cell lines in order to screen for new tumor related and metastatic candidate genes of nasopharyngeal carcinoma. METHODS: Three subtypes of nasopharyngeal carcinoma cell line SUNE 1 were analyzed by expression microarrays containing 14 000 cDNA clones, namely, the high metastatic potential cell line 5 8F, the tumorigenic non-metastatic line 6 10B and the non-tumorigenic line 13 9B MRNA expression was confirmed by semi-quantitative RT-PCR cDNA microarray test results. Results There were 82 differentially expressed genes between 5 8F and 13 9B. There were 38 differentially expressed genes between 6 10B and 13 9B. There were 54 differentially expressed genes between 5 8F and 6 10B, and 5 8F and 6 10B Compared with 13 9B and 12 9B, there were 12 common differentially expressed genes, while 5 8F had 14 common differentially expressed genes compared with 13 9B and 6 10B. These genes are involved in many biological functions such as metabolism, transcription, differentiation, apoptosis and signal transduction as well as many unknown functions. Conclusions revealed the gene expression profiles of nasopharyngeal carcinoma cell lines, providing an important clue for finding relevant genes of nasopharyngeal carcinoma
