目的:以药效实验优化回乳抑增方,用量效关系解释其最佳药味组成。方法:利用高泌乳素血症大鼠模型,再次验证最佳组方对泌乳素、雌激素和孕激素的调节作用,应用HPLC法测定各拆方中6种药效成分含量,比较各拆方药理作用优劣与方中主要药效物质含量差异的关联。结果:与模型组比较,原方与优化方均可使泌乳素(PRL)明显降低,雌二醇(E2)、孕酮(P)显著升高,其中对PRL、E2、P的改变作用以优化方最为明显(P<0.01)。优化方中的玄参、浙贝母在不影响迷迭香酸含量的同时还能保证大麦芽碱的溶出,药味之间存在增效现象。而其他各组药味配伍均能影响君药中大麦芽碱和迷迭香酸的溶出。结论:优化方药味组成合理,方中主要药效物质含量较大,药效作用最佳。
OBJECTIVE: To optimize the regimen of Yifan and Zifan by pharmacodynamics experiment, and explain the best composition of yogurt by dose-effect relationship. Methods: The model of hyperprolactinemia was used to verify the regulation of prolactin, estrogen and progesterone in the best prescription group. The contents of six effective components in each prescription were determined by HPLC. Pharmacological effects of pros and cons of the main side effects of the drug substance content of the correlation. Results: Compared with the model group, both prolactin and optimized prescription could significantly reduce the prolactin (PRL), estradiol (E2) and progesterone (P), and the changes of PRL, E2 and P Optimizer was the most obvious (P <0.01). Scrophulariaceae side, Fritillaria cirrhosa Maxim does not affect the rosmarinic acid content, but also ensure the dissolution of barley malt, there is synergy between the flavor. While the other groups can affect the compatibility of the drug king of barley malt and rosmarinic acid dissolution. CONCLUSION: The prescription of the optimized recipe has a reasonable composition, the major pharmacodynamic substances in the recipe are larger, and the best pharmacological effect is achieved.
