目的:研究大承气汤在不同配伍方法中枳实黄酮类成分的变化规律。方法:以君药大黄为核心,依次配伍其他3味中药(厚朴、枳实和芒硝),进行不同配伍方法设计,共得到7种配伍组方。采用HPLC法测定不同配伍组中柚皮苷、橙皮苷的含量。结果:大承气汤中枳实黄酮类成分在配伍环境中发生变化,经典煎法的柚皮苷和橙皮苷含量最高,分别为4.45%和1.16%。柚皮苷在0.168 5~1.164 8μg有良好的线性关系(r=0.999 5),平均回收率为98.20%,RSD为2.26%。结论:大承气汤全方配伍后,在一定程度上能增加黄酮类成分含量的溶出率。所建立的柚皮苷、橙皮苷HPLC含量测定方法具有较好的专属性,可用于本方质量控制和配伍指标的测定。
Objective: To study the changes of flavonoids in Danshen Fructus by different combinations of Dachengqi Decoction. Methods: The rhubarb was taken as the core, followed by compatibility with other three herbs (Magnolia officinalis, Citrus aurantium and Glauber’s salt). Different combinations of methods were designed and seven kinds of compatibility prescriptions were obtained. The contents of naringin and hesperidin in different compatibility groups were determined by HPLC. Results: The contents of flavonoids in Fructus Aurantii Immaturus in Dachengqi Decoction changed in compatibility environment. The contents of naringin and hesperidin in classic decoction were the highest, which were 4.45% and 1.16% respectively. Naringin in 0.168 5 ~ 1.164 8μg has a good linear relationship (r = 0.999 5), the average recovery was 98.20%, RSD was 2.26%. Conclusion: Dachengqi Decoction compatibility, to a certain extent, can increase the dissolution rate of flavonoid content. The established method for the determination of naringin and hesperidin HPLC has good specificity and can be used for the quality control of the side and the determination of compatibility index.
