聚合酶链式反应(Polymerase Chain Reaction),简称PCR,是一种分子生物学技术,用于放大特定的DNA片段,可看作生物体外的特殊DNA复制。随着分子生物学技术渗透到生物学科的各个领域,领路者“PCR”技术也在中学生物教材中跃然纸上。一直以来,中学开展PCR实验的教学效果都不尽理想。本文主要从引物设计,PCR反应体系的优化以及Tm值的合理选择几个方面谈谈对改善中学PCR实验的几点思考,同时还对防治实验结果的污染及检测进行了阐述。
Polymerase Chain Reaction, or PCR for short, is a molecular biology technique that amplifies specific DNA fragments and can be thought of as a special DNA replication in vitro. With the penetration of molecular biology technology into all fields of biology, the leader of “PCR” technology is vividly printed on middle school biology textbooks. All along, teaching high school PCR experiments are not satisfactory. This article mainly from the primer design, PCR reaction system optimization and Tm value of the rational choice of several aspects to improve the high school PCR experiment thinking, but also on the prevention and treatment of pollution and detection results are described.
