用体外全胚胎培养(wholeembryoculture,WEC)探讨硒、氟和砷三者以不同配比浓度对旋转培养48h的大鼠胚胎所产生的联合作用。3×3×3析因分析结果表明硒、氟、砷联合对发育呈协同毒性作用;随着三种化学物剂量的配比变化,发育毒性效应亦不同,7个低剂量联合组的协同作用主要表现为致畸性,而高剂量联合组〔(2.0μg硒+10.0μg氟+1.0μg砷)/ml培养基〕的协同作用以致死性为主。结果还提示卵黄囊胎盘结构与功能紊乱是硒、氟、砷联合致畸的重要机制之一。
In vitro whole embryo culture (whole embryo culture) was used to investigate the combined effects of selenium, fluoride and arsenic at different concentrations on rat embryos cultured for 48 h. The result of factorial analysis of 3 × 3 × 3 showed synergistic toxicity of selenium, fluorine and arsenic in development. With the change of the ratio of the three chemicals, the developmental toxicity effect was also different. The synergistic effects of the seven low-dose combination groups The main manifestation of teratogenicity, and high-dose combination group 〔(2.0μg of selenium + 10.0μg fluoride + 1.0μg arsenate) / ml medium〕 synergistic to lethal. The results also suggest that the structure and function of yolk sac placenta is one of the important mechanisms of teratogenicity combined with selenium, fluorine and arsenic.
