目的 探讨蛋白酪氨酸磷酸酶 1B(proteintyrosinephosphastase 1B ,PTP 1B)基因 387位编码子Pro Leu多态性与 2型糖尿病 (T2DM )的关系。方法 采用多聚酶联反应 限制性片段长度多态性 (PCR RFLP)的方法对湖北地区 1 30例 2型糖尿病患者及 1 38例正常对照者PTP 1B基因 387位编码子酶切位点进行研究。结果 2型糖尿病患者和正常对照者PTP 1B基因均以PP基因型为主 ,其频率分别为 0 .94和 0 .96 ;P等位基因频率分别为 0 .97和 0 .98,差异无统计学意义 (P >0 .0 5)。 2型糖尿病患者中PTP 1B基因P387L变异与体重指数 (BMI)有关 ,但与空腹血糖、空腹胰岛素、总胆固醇、甘油三酯等临床变量不相关。结论 未发现PTP 1B基因 387位Pro Leu多态性与中国人 2型糖尿病有关
Objective To investigate the relationship between the 387-coding pro-Leu polymorphism of protein tyrosine phosphastase 1B (PTP 1B) gene and type 2 diabetes mellitus (T2DM). Methods The 387-coding site of PTP 1B gene in 130 type 2 diabetic patients and 138 healthy controls in Hubei Province was studied by polymerase chain reaction restriction fragment length polymorphism (PCR RFLP). Results The genotypes of PTP 1B in patients with type 2 diabetes mellitus and normal controls were PP genotypes with frequencies of 0.94 and 0.96, respectively. The frequencies of P alleles were 0.97 and 0.98, respectively, with no significant difference Significance of learning (P> 0.05). The PTP 1B gene P387L variation in type 2 diabetic patients is associated with body mass index (BMI) but not with clinical variables such as fasting plasma glucose, fasting insulin, total cholesterol, and triglycerides. Conclusion The 387-bp Pro Leu polymorphism of PTP 1B gene was not found to be associated with type 2 diabetes in Chinese
