目的:研究临床常用的窝沟封闭剂Densply的体外细胞毒性。方法:用细胞培养液浸提窝沟封闭剂Den-sply24h,配置不同浓度的浸提液(0.1~100 mg/mL),将小鼠成纤维细胞(L-929)在不同浓度的窝沟封闭剂Densply浸提液中分别培养24 h,通过Alamar Blue比色法,检测窝沟封闭剂Densply对L-929细胞相对增殖率(RGR)的影响,评价其细胞毒性。结果:L-929细胞呈现高的细胞增殖率,各浓度窝沟封闭剂Densply浸提液之间差异无显著性意义(P>0.05),其细胞毒性为0~1级。结论:高分子交联聚合物聚磷酸酯无明显的细胞毒性,具有较好的生物相容性。
Objective: To study the in vitro cytotoxicity of Densply, a commonly used pit and fissure sealant. Methods: The fissure sealant Den-sply24h was digested with cell culture solution and different concentrations of leaching solution (0.1-100 mg / mL) were prepared. Mouse fibroblasts (L-929) Densply extracts were incubated for 24 h respectively. The effects of pit and fissure sealant Densply on the relative proliferation rate (RGR) of L-929 cells were detected by Alamar Blue colorimetric assay to evaluate the cytotoxicity. Results: L-929 cells showed high cell proliferation rate. There was no significant difference (P> 0.05) between Densply extracts of pit and fissure sealant. The cytotoxicity was 0 ~ 1 grade. Conclusion: The polymer cross-linked polymer polyphosphate has no obvious cytotoxicity and has good biocompatibility.
