胃痞消对脾虚型慢性萎缩性胃炎癌前病变大鼠胃黏膜上皮细胞增殖周期分布及凋亡基因表达的影响

【目的】观察健脾化瘀解毒中药胃痞消(由黄芪、太子参、白术、丹参、白花蛇舌草等组成)对脾虚型慢性萎缩性胃炎(CAG)癌前病变大鼠胃黏膜上皮细胞的增殖周期分布及凋亡基因表达的影响。【方法】选用SD大鼠,随机分为6组,即正常对照组,模型组,胃痞消高、中、低剂量组(剂量分别为15、7.5、3.75g·kg-1·d-1),维酶素组(剂量为0.27g·kg-1·d-1)。除正常对照组外,其他组均采用N-甲基-N’-硝基-N-亚硝基胍(MNNG)液自由饮用结合饥饱失常加耗气泻下法复制脾虚型CAG胃癌前病变模型,观察胃痞消各剂量组连续治疗4周后对大鼠胃黏膜上皮细胞增殖周期分布、凋亡率、凋亡基因p53、Bcl-2和Bax表达的影响。【结果】胃痞消各剂量组可显著增加胃黏膜上皮细胞凋亡率,增加G0/G1期、减少S期细胞分布,促进p53表达,抑制Bcl-2表达(均P<0.01),胃痞消高剂量组可显著促进Bax表达(P<0.05)。【结论】胃痞消可抑制CAG癌前病变大鼠胃黏膜上皮细胞的增殖,促进细胞凋亡,其机制可能与其能促进p53和Bax表达,抑制Bcl-2表达有关。 【Objective】 To observe the effect of Weipinghuxujiedu Decoction (consisting of astragalus, heterophylla, Atractylodes, Salvia miltiorrhiza, Hedyotis diffusa) on gastric mucosal epithelial cells of rat with chronic atrophic gastritis (CAG) precancerous lesion Proliferation cycle distribution and apoptosis gene expression. 【Methods】 SD rats were randomly divided into 6 groups: normal control group, model group, Weipixiao high, medium and low dose groups (doses of 15, 7.5 and 3.75 g · kg -1 · d -1 ), The vitamin group (dose of 0.27g · kg-1 · d-1). In addition to the normal control group, the other groups were treated with N-methyl-N’-nitro-N-nitrosoguanidine (MNNG) Model to observe the effects of Weipixiao Decoction for four weeks on the distribution of gastric epithelial cell proliferation, the apoptotic rate and the expressions of p53, Bcl-2 and Bax in gastric mucosal epithelial cells. 【Results】 Weibai consumption of each dose group could significantly increase the apoptosis rate of gastric mucosal epithelial cells, increase G0 / G1 phase, reduce the S phase cell distribution, promote p53 expression, inhibit the expression of Bcl-2 (all P <0.01) Xiaogao dose group can significantly promote the expression of Bax (P <0.05). 【Conclusion】 Weipixiao can inhibit the proliferation and promote the apoptosis of gastric epithelial cells in CAG precancerous lesion rats. The mechanism may be related to the promotion of p53 and Bax expression and the inhibition of Bcl-2 expression.
