目的研究食欲素受体1(OX1R)、食欲素受体2(OX2R)、抗中肾旁管激素受体Ⅱ(AMHR2)在下丘脑-垂体-睾丸轴中分布与细胞定位,探讨其对生精功能的调节机制。方法用免疫组织化学法检测OX1R,OX2R和AMHR2受体在性未成熟和性成熟SD大鼠的下丘脑、垂体、睾丸表达情况。结果性未成熟组:OX1R,OX2R和AMHR2受体在下丘脑室周带、内侧带、外侧带神经元细胞质及细胞膜表达阳性,在腺垂体少数细胞细胞质阳性,在睾丸只有间质细胞细胞膜及细胞质表达阳性。性成熟组:OX1R、OX2R和AMHR2受体在下丘脑室周带、内侧带、外侧带神经元细胞质及细胞膜表达阳性,在腺垂体少数细胞细胞质阳性,在睾丸间质细胞和精原细胞细胞膜及细胞质表达阳性。结论 OX1R、OX2R和AMHR2受体在性成熟与未成熟的大鼠下丘脑、脑垂体表述无差异,在睾丸分布上具有不同,上述3种受体可能参与性腺轴调节精原细胞分化功能。
Objective To investigate the distribution and location of OX1R, OXR2 and AMHR2 in the hypothalamus-pituitary-testis axis and to investigate the effects of OX1R, OXR2, Function adjustment mechanism. Methods Immunohistochemistry was used to detect the expression of OX1R, OX2R and AMHR2 receptors in the hypothalamus, pituitary and testis of sexual immature and sexually mature SD rats. Results immature group: OX1R, OX2R and AMHR2 receptors in the hypothalamic ventricular zone, medial and lateral neuronal cytoplasm and cell membrane expression was positive in the pituitary gland a small number of cells in the cytoplasm of the testis, only the stromal cells in the plasma membrane and cytoplasm Positive. Sexual maturation group: OX1R, OX2R and AMHR2 receptors were positive in the cytoplasm and cell membrane of the periventricular zone, medial zone and lateral zone of the hypothalamus. A few cells in the pituitary gland were cytoplasm positive. In the testicular stromal cells and spermatogonial cell membrane and cytoplasm Positive expression. Conclusion OX1R, OX2R and AMHR2 receptors have no difference in hypothalamus and pituitary expression in mature and immature rats, but different in testis distribution. The above three receptors may participate in the gonadal axis regulating the differentiation of spermatogonia.
