目的:通过观察重组白介素-12治疗小鼠病毒性心肌炎过程中干扰素-γ及白介素-10水平的变化,探究干扰素-γ及白介素-10在重组白介素-12对病毒性心肌炎治疗中的作用。方法:近交BALB/C小鼠140只,雄性,随机分成6组,空白对照组,病毒对照组,药物对照组,小剂量治疗组,中剂量治疗组和大剂量治疗组,除空白对照组外,每组小鼠腹腔注射柯萨奇病毒B_3共3天,药物治疗连续5天。于实验第5、10、15天每组分别取4只鼠,摘眼球取血应用ELISA法检测血清干扰素-γ及白介素-10水平。取血后拉颈处死,摘取心脏,福尔马林固定,用于做心肌病理学检测。结果:不同治疗组之间和治疗后的时间对干扰素-γ及白介素-10值均有影响。随着时间变化的比较,第5天除空白对照组外,各组心肌炎症细胞浸润最重,第10天开始恢复,第15天心肌间质纤维母细胞增多,处于恢复期。各组间比较,中剂量治疗组病变最轻,大剂量治疗组病变最重,而小剂量治疗组和药物对照组病变接近,但轻于大剂量治疗组。结论:重组白介素-12对病毒性心肌炎小鼠有治疗作用,以10 ng/只的剂量治疗效果最好。干扰素-γ及白介素-10在重组白介素-12的治疗中起一定的作用。
OBJECTIVE: To investigate the effects of IFN-γ and IL-10 on the treatment of viral myocarditis by recombinant interleukin-12 (IFN-γ) and interleukin-10 (IL-10) during viral myocarditis in mice . Methods: 140 male BALB / C mice were randomly divided into 6 groups, blank control group, virus control group, drug control group, low dose treatment group, middle dose treatment group and high dose treatment group, except for blank control group In addition, Coxsackie virus B_3 was intraperitoneally injected into the mice in each group for 3 days for 5 consecutive days. On the 5th, 10th, and 15th day, 4 mice were taken from each group, and blood was taken from the eyeballs. Serum levels of interferon-γ and interleukin-10 were measured by ELISA. Take the neck after death, remove the heart, formalin fixed, used for myocardial pathology. Results: Interferon-γ and interleukin-10 values were influenced by different treatment groups and after treatment. With the change of time, on the 5th day except for the blank control group, the infiltration of myocardial inflammatory cells in each group was the heaviest, and began to recover on the 10th day. The myocardial interstitial fibroblasts increased on the 15th day and were in recovery. Compared with each group, the lesion in the middle-dose treatment group was the lightest, the lesion in the high-dose treatment group was the heaviest, while the lesions in the low-dose treatment group and the drug control group were close, but lighter than the high-dose treatment group. CONCLUSION: Recombinant IL-12 has a therapeutic effect on mice with viral myocarditis, and the best dose of 10ng / Interferon-γ and interleukin-10 play a role in the treatment of recombinant interleukin-12.
