目的探讨肺癌细胞中膜骨架连接蛋白Ezrin对体外培养的肺癌细胞系的增殖、凋亡、侵袭和转移能力的影响,明确Ezrin在肺癌中的异常表达与临床病理因素之间的关系。方法转染EzrinSiRNA后,应用流式、MTT、Transwell和划痕实验等技术评价肺癌细胞体外侵袭转移和增殖能力的变化。结果转染EzrinSiRNA后,划痕实验、MTT和Transwell等实验结果证明肺癌细胞的侵袭和增殖能力减弱。流式细胞仪技术检测凋亡的结果证明Ezrin沉默后,肺癌细胞凋亡数目增加。结论膜细胞骨架连接蛋白Ezrin可以明显促进肺癌细胞的侵袭转移。
Objective To investigate the effect of Ezrin on the proliferation, apoptosis, invasion and metastasis of lung cancer cell lines in vitro and to elucidate the relationship between the abnormal expression of Ezrin and clinicopathological factors in lung cancer. Methods After transfection of EzrinSiRNA, the invasion, metastasis and proliferation of lung cancer cells were evaluated by flow cytometry, MTT, Transwell and scratch assay. Results After transfection of EzrinSiRNA, scratch test, MTT and Transwell experiments showed that invasion and proliferation of lung cancer cells were weakened. The results of flow cytometry to detect apoptosis showed that the number of apoptotic lung cancer cells increased after Ezrin silencing. Conclusion Membrane cytoskeleton connexin Ezrin can significantly promote the invasion and metastasis of lung cancer cells.
