目的:观察骨髓间充质干细胞(BMSC)植入猪急性心肌梗死(AMI)区的转归及治疗作用。方法:选择10只体重35~45kg的家猪,经导管应用球囊封堵前降支动脉,建立心肌梗死模型,将存活的9只动物随机分为2组:(1)实验组5只,取骨髓分离BMSC体外培养扩增,建立模型2周后,通过心肌注射的方法将自体骨髓BMSC移植至AMI区;(2)对照组4只动物注射等量生理盐水。4周后比较2组的生存率、心功能变化、心肌功能性蛋白质表达的差异。结果:BMSC移植后,实验组动物心功能得到明显改善,梗死区室壁运动速度加快,局部室壁厚度加快,心肌移植细胞部位表达连接蛋白GATA-4等心肌功能性蛋白质。结论:骨髓BMSC经冠脉转运在心肌微环境下可向心肌细胞分化,并能改善心功能。
Objective: To observe the prognosis and therapeutic effect of bone marrow mesenchymal stem cells (BMSCs) implanted in pig acute myocardial infarction (AMI). Methods: Ten domestic pigs weighing 35-45 kg were selected. The anterior descending artery was occluded by balloon catheterization and the myocardial infarction model was established. Nine surviving animals were randomly divided into two groups: (1) experimental group (5) BMSCs were isolated and cultured in vitro. After 2 weeks, the BMSCs were transplanted into the AMI by intramyocardial injection. (2) Four animals in the control group were injected with the same amount of saline. Four weeks later, the difference of survival rate, cardiac function and myocardial functional protein expression in two groups were compared. RESULTS: After transplanted with BMSC, the heart function of the experimental group was significantly improved. The wall motion of the infarct area was accelerated and the thickness of the local wall was accelerated. Cardiac function proteins such as connexin GATA-4 were expressed in the transplanted cells. CONCLUSION: BMSCs transdifferentiated by coronary artery can differentiate into cardiomyocytes in myocardial microenvironment and improve cardiac function.
