目的:观察活血养血汤对兔脊髓缺血再灌注损伤Cytc、Hsp70、Bcl-2、Bax表达的影响,探讨其作用机制。方法:新西兰大白兔随机分成手术假手术组、手术组、药物对照组和活血养血汤组,每组30只,术前1周分别灌服0.9%氯化钠溶液、桃红四物汤和活血养血汤,假手术组只分离出腰动脉,其余3组制作脊髓缺血再灌注损伤模型,4组分别于缺血再灌注后0.5、1、4、8、12h检测Cytc、Hsp70、Bcl-2、Bax表达。结果:药物对照组和活血养血汤组各时间点Cytc表达较手术组有显著下降(P<0.05,P<0.01),活血养血汤组较药物对照组各时间点相比有显著下降(P<0.05,P<0.01);药物对照组与活血养血汤组Hsp70和Bcl-2表达在4、8、12h较手术组明显升高(P<0.05,P<0.01),8、12h活血养血汤组较药物对照组显著升高(P<0.05,P<0.01);药物对照组和活血养血汤组Bax表达各时间点均低于手术组(P<0.05,P<0.01),且活血养血汤组低于药物对照组(P<0.05,P<0.01)。结论:桃红四物汤和活血养血汤可通过降低Cytc、Bax,升高Hsp70、Bcl-2的表达调控细胞凋亡,减轻损伤,活血养血汤后期作用优于单纯活血化瘀法。
Objective: To observe the effect of Huoxue Yangxue Decoction on the expression of Cytc, Hsp70, Bcl-2 and Bax after spinal cord ischemia-reperfusion injury in rabbits and to explore its mechanism. Methods: New Zealand white rabbits were randomly divided into operation sham operation group, operation group, drug control group and Huoxue Yangxue Decoction group, 30 rats in each group. 0.9% sodium chloride solution, Taohong Siwu Decoction and blood circulation Yangxue Decoction and sham operation group, only the lumbar artery was isolated, and the other three groups were made into the model of spinal cord ischemia-reperfusion injury. The four groups were respectively detected Cytc, Hsp70 and Bcl- 2, Bax expression. Results: The expression of Cytc in drug control group and Huoxue Yangxue Decoction group were significantly lower than those in operation group (P <0.05, P <0.01), and Huoxue Yangxue Decoction group was significantly lower than that in drug control group at each time point P <0.05, P <0.01). The expression of Hsp70 and Bcl-2 in the drug control group and Huoxue Yangxue Decoction group were significantly higher than those in the operation group at 4,8 and 12 h (P <0.05, P <0.01) Yangxue Decoction group was significantly higher than the drug control group (P <0.05, P <0.01); Bax expression of the drug control group and Huoxue Yangxue Decoction group were lower than those of the operation group at each time point (P <0.05, P <0.01) And Huoxue Yangxue Decoction group was lower than the drug control group (P <0.05, P <0.01). Conclusion: Taohong Siwu Decoction and Huoxue Yangxue Decoction can regulate apoptosis and reduce injury by decreasing the expression of Cytc, Bax, increasing the expression of Hsp70 and Bcl-2. The effect of Huoxue Yangxue Decoction is better than that of promoting blood circulation and removing blood stasis alone.
