目的①探讨年龄、性别对封闭群小鼠(KM、ICR)不同脑区Munc18-1含量的影响;②探讨年龄、性别对封闭群小鼠(KM、ICR)血清甲状腺激素水平的影响;③探讨Munc18-1含量与血清甲状腺激素水平之间的相关性。方法①选取3个年龄段(22月龄老年组、11月龄中年组、6月龄青年组)昆明小鼠和2个年龄段(12月龄中年组、7月龄青年组)ICR小鼠;②应用Western blot检测小鼠嗅球、额叶、顶叶、腹侧海马及背侧海马中Munc18-1和内参蛋白β-actin蛋白的表达。Munc18-1灰度值与β-actin灰度值的比值作为Munc18-1相对含量;②采用放射免疫法检测小鼠血清甲状腺激素水平;③用方差分析、Spearman秩相关检验进行统计学分析。结果①22月龄昆明小鼠背侧海马Munc18-1含量显著高于中年鼠和青年鼠(P<0.05),且以老年雌鼠显著(P<0.05),而中年昆明小鼠与青年鼠之间未发现显著性差异(P>0.05);12月龄ICR小鼠背侧海马Munc18-1相对含量显著高于青年鼠(P<0.05),且以雌鼠显著(P<0.05)。②22月龄昆明小鼠血清FT3、FT4水平显著低于青年鼠(P<0.05),且FT3显著低于中年鼠(P<0.05),以雌鼠显著(P<0.05),而中年与青年鼠之间未发现显著性差异(P>0.05);12月龄ICR小鼠血清FT3水平显著低于青年鼠(P<0.05),以雌鼠显著(P<0.05)。③昆明小鼠和ICR小鼠背侧海马Munc18-1含量均与FT3呈负相关(P<0.05)。结论①封闭群小鼠背侧海马Munc18-1含量可出现年龄相关性显著升高,其中以雌鼠升高更明显;②封闭群小鼠可出现年龄相关性血清甲状腺激素水平显著降低,其中以雌鼠降低更明显;③血清甲状腺激素水平降低可能是导致背侧海马Munc18-1含量升高的原因之一。
Objectives ① To investigate the effects of age and gender on Munc18-1 levels in different brain regions of closed mice (KM, ICR); ② to investigate the effects of age and gender on serum thyroid hormones in closed mice (ICR); ③ to explore Correlation between Munc18-1 levels and serum thyroid hormone levels. Methods ① Kunming mice of three age groups (22 months old group, 11 months middle age group, 6 months old group) and 2 age groups (12 months old middle-aged group, 7-month old young group) Mice; ② Western blot was used to detect the expression of Munc18-1 and β-actin protein in the olfactory bulb, frontal lobe, parietal lobe, ventral hippocampus and dorsal hippocampus. Munc18-1 gray value and β-actin grayscale value as the relative content of Munc18-1; ② using radioimmunoassay serum thyroid hormone levels; ③analysis of variance, Spearman rank correlation test for statistical analysis. Results ① The content of Munc18-1 in the dorsal hippocampus of 22-month-old Kunming mice was significantly higher than that of middle-aged rats and young rats (P <0.05), and was significantly higher in aged females (P <0.05) (P> 0.05). The relative content of Munc18-1 in the dorsal hippocampus of 12-month-old ICR mice was significantly higher than that of the young mice (P <0.05). (2) The levels of FT3 and FT4 in 22-month-old Kunming mice were significantly lower than those in young mice (P <0.05), FT3 was significantly lower than that in middle-aged mice (P <0.05) There was no significant difference between young mice (P> 0.05). The serum FT3 levels in 12-month-old ICR mice were significantly lower than those in young mice (P <0.05), and significantly in female mice (P <0.05). ③ The content of Munc18-1 in Kunming mice and ICR mice dorsal hippocampus were negatively correlated with FT3 (P <0.05). Conclusion ① The Munc18-1 content in the dorsal hippocampus of the mice in the closed group may appear age-related increase significantly, especially in the females; ② The age-related serum thyroid hormone levels may be significantly decreased in the closed mice, Female rats reduce more obvious; ③ serum thyroid hormone levels may be caused by dorsal hippocampus Munc18-1 content increased one of the reasons.
