目的:探讨蓝萼甲素对人脐静脉内皮细胞(HUVECs)的保护作用。方法:0.1 mmol/L过氧化氢(H2O2)作用于人脐静脉内皮细胞株,实验分为空白对照组、氧化损伤模型组、阿司匹林对照组和蓝萼甲素低、中、高剂量组。RT-PCR检测内皮细胞ET-1、iNOS、eNOS的表达。结果:与模型组比较,蓝萼甲素低、中、高剂量组均能升高细胞增殖率,蓝萼甲素中、高剂量组ET-1和iNOS表达明显减少,eNOS表达明显增多。结论:蓝萼甲素能够明显改善H2O2对内皮细胞的损伤,可能是通过降低ET-1而减少血管的收缩;降低iNOS表达而减少内皮细胞的损伤,达到保护内皮细胞的作用。
Objective: To investigate the protective effect of CVE on human umbilical vein endothelial cells (HUVECs). Methods: Human umbilical vein endothelial cell line was induced by 0.1 mmol / L hydrogen peroxide (H2O2). The experiment was divided into blank control group, oxidative injury model group, aspirin control group and hydrocortisone A1 low, medium and high dose groups. The expression of ET-1, iNOS and eNOS in endothelial cells was detected by RT-PCR. Results: Compared with the model group, the cell proliferation rate was increased in the low, medium and high dose group of the blue callactone, while the expression of ET-1 and iNOS was significantly decreased and the eNOS expression was significantly increased. CONCLUSION: Clarithromycin A can significantly improve the damage of endothelial cells induced by H2O2, which may be due to the decrease of ET-1 and the reduction of vasoconstriction; reduce the expression of iNOS and reduce the damage of endothelial cells and thus protect the endothelial cells.
