目的研究CDC25B基因在胰腺癌组织中的表达,探讨其在胰腺癌发病机制中的作用。方法利用实时荧光定量-PCR技术检测24例胰腺癌组织和4例正常胰腺组织中CDC25B基因的表达,PCR产物的定量方法采用比较Ct法,应用western blot检测其蛋白的表达水平。用Kaplan-Meier生存率曲线分析CDC25BmRNA与预后的关系。结果实时荧光定量-PCR发现CDC25B基因平均模板量在正常胰腺组织中为0.000635±0.000210,胰腺癌组织中为0.001988±0.000650;与正常胰腺组织相比,胰腺癌组织中CDC25B mRNA表达上调3.13倍(P<0.05),CDC25B mRNA升高5倍以上者生存时间缩短。胰腺癌组织中CDC25B蛋白也过度表达,正常胰腺组织CDC25B蛋白表达为1.22±0.33,胰腺癌组织为5.56±1.57,上调4.56倍(P<0.01)。结论CDC25B在胰腺癌组织中表达明显上调,CDC25B mRNA升高者预后差。
Objective To study the expression of CDC25B gene in pancreatic cancer and its role in the pathogenesis of pancreatic cancer. Methods The expression of CDC25B gene in 24 cases of pancreatic cancer and 4 cases of normal pancreas was detected by real-time fluorescence quantitative-PCR. The quantitative method of PCR products was compared with Ct method and the protein expression level was detected by western blot. The relationship between CDC25B mRNA and prognosis was analyzed by Kaplan-Meier survival curve. Results The average amount of CDC25B gene was 0.000635 ± 0.000210 in normal pancreatic tissues and 0.001988 ± 0.000650 in pancreatic cancer tissues by real-time fluorescence quantitative PCR. Compared with normal pancreatic tissues, CDC25B mRNA was up-regulated by 3.13-fold (P <0.05), CDC25B mRNA increased more than 5 times shorter survival time. The expression of CDC25B protein in pancreatic cancer tissues was also overexpressed. The expression of CDC25B protein in normal pancreatic tissues was 1.22 ± 0.33 and in pancreatic cancer tissues was 5.56 ± 1.57, up-regulated by 4.56-fold (P <0.01). Conclusion The expression of CDC25B in pancreatic cancer tissues was significantly up-regulated, and the prognosis of patients with elevated CDC25B mRNA was poor.
