目的探讨不同浓度葡萄糖对INS-1细胞PTHrP/PTH1R表达的影响以及对细胞增殖的作用。方法将大鼠胰岛细胞瘤细胞株根据不同干预条件分为空白对照组,正常葡萄糖组(5mmol/L)组及高浓度葡萄糖(30mmol/L)组,培养96h应用MTT检测各组细胞活力,应用Western blot及RT-PCR检测PTHrP/PTH1R蛋白及mRNA表达。结果空白对照组及高浓度葡萄糖组细胞活力比正常葡萄糖组下降,高浓度葡萄糖组细胞活力低于无糖组,PTHrP和PTH1R蛋白或mRNA在高浓度葡萄糖组中表达显著下调。结论不同浓度葡萄糖可以通过调控PTHrP/PTH1R的水平从而影响INS-1的增殖。
Objective To investigate the effects of different concentrations of glucose on the expression of PTHrP / PTH1R in INS-1 cells and its effect on cell proliferation. Methods Rat pancreatic islet cell lines were divided into blank control group, normal glucose group (5mmol / L) and high concentration glucose (30mmol / L) group according to different intervention conditions. After cultured for 96h, cell viability was measured by MTT assay. The expressions of PTHrP / PTH1R protein and mRNA were detected by Western blot and RT-PCR. Results The viability of blank control group and high concentration glucose group was lower than that of normal glucose group. The cell viability of high concentration glucose group was lower than that of non - glucose group. The expression of PTHrP and PTH1R protein or mRNA was significantly down - regulated in high glucose group. Conclusion Different concentrations of glucose can affect the proliferation of INS-1 by regulating the level of PTHrP / PTH1R.
