牛磺酸对成纤维细胞增殖及功能的影响

目的:观察牛磺酸对3T3细胞的增殖及其产生胶原和透明质酸的影响。方法:采用[3H]-胸腺嘧啶([3H]-TdR)、[3H]-脯氨酸([3H]-Pro)掺入和放射免疫分析法分别测定细胞增殖、胶原和透明质酸合成。结果:牛磺酸能显著抑制3T3细胞的[3H]-TdR和[3H]-Pro掺入,并呈时间、剂量依赖关系;对透明质酸的合成亦具有剂量依赖的抑制作用。此外,牛磺酸还明显提高细胞内cAMP水平。结论:牛磺酸能显著抑制3T3细胞增殖及胶原和透明质酸合成,其机理可能与促进cAMP生成有关。 Objective: To observe the effect of taurine on the proliferation of 3T3 cells and the production of collagen and hyaluronic acid. METHODS: Cell proliferation, collagen, and hyaluronan synthesis were determined using [3H]-thymidine ([3H]-TdR), [3H]-proline ([3H]-Pro) incorporation, and radioimmunoassay, respectively. RESULTS: Taurine could significantly inhibit the incorporation of [3H]-TdR and [3H]-Pro in 3T3 cells in a time- and dose-dependent manner. It also had a dose-dependent inhibitory effect on the synthesis of hyaluronic acid. In addition, taurine significantly increased intracellular cAMP levels. CONCLUSION: Taurine can significantly inhibit the proliferation of 3T3 cells and the synthesis of collagen and hyaluronan, and its mechanism may be related to the promotion of cAMP production.
