目的:建立高效液相色谱法测定小金丸中乌头碱的含量。方法:采用ultimate C18柱(4.6 mm×250 mm,5μm),以甲醇-0.03 mol.L-1磷酸二氢钾溶液-三乙胺(45∶55∶0.04)为流动相,流速1.0 mL.min-1,检测波长240 nm,柱温25℃。结果:乌头碱在0.114~2.28μg具有良好的线性关系(r=0.999 8),平均回收率为97.79%,RSD 2.31%。结论:该方法准确、灵敏,重复性好,可用于小金丸中乌头碱的含量测定。
Objective: To establish a HPLC method for the determination of aconitine in Xiaojin Pills. Methods: The optimal C18 column (4.6 mm × 250 mm, 5 μm) was used with methanol-0.03 mol·L-1 potassium dihydrogen phosphate solution-triethylamine (45:55:0.04) as the mobile phase at a flow rate of 1.0 mL · min -1, detection wavelength 240 nm, column temperature 25 ℃. Results: Aconitine had a good linearity (r = 0.999 8) between 0.114 and 2.28 μg with an average recovery of 97.79% and a RSD of 2.31%. Conclusion: The method is accurate, sensitive and reproducible. It can be used for the determination of aconitine in Xiaojin Pills.
