目的:观察实验性创伤性骨关节炎大鼠软骨和滑膜组织中NF-κBp65的表达。方法:雄性SD大鼠24只,随机分为对照组(n=12)和造模组(n=12),造模组采用切断膝关节内侧副韧带和部分切除内侧半月板建立大鼠创伤性骨关节炎模型。两周后处死大鼠,采用蛋白质免疫印迹法(WesternBlot)和免疫组织化学染色法检测大鼠关节软骨和滑膜内的NF-κBp65表达。结果:与对照组相比,造模组关节软骨和滑膜中的NF-κBp65表达显著增高。结论:NF-κBp65可能在创伤性关节炎的发生过程中发挥作用。
Objective: To observe the expression of NF-κBp65 in cartilage and synovial tissue of experimental traumatic osteoarthritis in rats. Methods: Twenty-four male Sprague-Dawley rats were randomly divided into control group (n = 12) and model group (n = 12). The model group was established by cutting the medial collateral ligament of knee and partial resection of the medial meniscus Osteoarthritis model. Two weeks later, the rats were sacrificed and the expression of NF-κBp65 in articular cartilage and synovium was detected by Western blotting and immunohistochemistry. Results: Compared with the control group, the expression of NF-κBp65 in the articular cartilage and synovium was significantly increased. Conclusion: NF-κBp65 may play a role in the pathogenesis of traumatic arthritis.
