目的比较蒽酮-硫酸法与苯酚-硫酸法测定不同切面颜色(红色、白色)土茯苓药材中多糖的含量差异,建立土茯苓中多糖含量测定方法。方法分别采用蒽酮-硫酸法与苯酚-硫酸法,在紫外可见分光光度计下测定切面红色、白色土茯苓药材中多糖的含量信息,并进行方法学考察,比较两种方法测定结果的差异。结果苯酚硫酸法含量测定结果高于蒽酮硫酸法,其差异有统计学意义,苯酚硫酸法的线性、重现性和稳定性均优于蒽酮硫酸法。结论苯酚硫酸法比蒽酮硫酸法更适合测定土茯苓中多糖的含量。
Objective To compare the contents of polysaccharides in different cross-section color (red and white) of Radix Scutellariae with flavonoids-sulfuric acid method and phenol-sulfuric acid method, and to establish a method for the determination of polysaccharides in Radix Scutellariae. Methods The contents of polysaccharides in red and white radix sculpina were determined by anthrone-sulfuric acid method and phenol-sulfuric acid method. The method was used to investigate the differences of the results of the two methods. Results The results of determination of phenol-sulfuric acid content were higher than that of anthrone-sulfate method. The difference was statistically significant. The linearity, reproducibility and stability of the phenol-sulfuric acid method were better than those of the anthrone-sulfate method. Conclusion The phenol-sulfuric acid method is more suitable than the anthrone-sulfuric acid method for the determination of polysaccharides in Smilax china.
