目的:建立同时测定左金丸乙醇提取物中表小檗碱、盐酸药根碱、黄连碱、盐酸巴马汀、盐酸小檗碱、吴茱萸碱和吴茱萸次碱含量的液相色谱法。方法:采用Kromasil C18(4.6 mm×250 mm,5μm)柱,以乙腈-0.1%磷酸和0.05%三乙胺水溶液为流动相进行梯度洗脱,检测波长0~40 min为265 nm,40~55 min为225 nm。结果:表小檗碱、盐酸药根碱、黄连碱、盐酸巴马汀、盐酸小檗碱、吴茱萸碱、吴茱萸次碱的线性范围分别是0.275 9~17.66,0.267 0~17.09,0.262 5~16.80,0.221 4~14.17,0.948 7~60.71,0.050 4~3.229,0.044 9~2.874 mg·L-1,平均回收率分别为100.4%,99.8%,100.2%,100.0%,99.0%,98.9%,97.4%,RSD分别为1.8%,1.2%,2.1%,2.1%,2.0%,2.0%,2.2%。结论:本法简便、准确,可同时测定左金丸乙醇提取物中表小檗碱、盐酸药根碱、黄连碱、盐酸巴马汀、盐酸小檗碱、吴茱萸碱和吴茱萸次碱的含量,为左金丸的现代制剂研究和质量控制提供方法学参考。
OBJECTIVE: To establish a liquid chromatography method for the simultaneous determination of berberine, jatrorrhizine hydrochloride, berberine hydrochloride, palmatine hydrochloride, berberine hydrochloride, evodiamine and rutaecarpine in Zuojin Pills ethanol extract. Methods: The gradient elution was carried out on a Kromasil C18 column (4.6 mm × 250 mm, 5 μm) using acetonitrile-0.1% phosphoric acid and 0.05% triethylamine as the mobile phase. The detection wavelength was set at 265 nm and 40-55 at 0-40 min min is 225 nm. Results: The linear ranges of berberine, jatrorrhizine hydrochloride, berberine hydrochloride, palmatine hydrochloride, berberine hydrochloride, evodiamine and rutaecarpine were 0.275 9 ~ 17.66, 0.267 0 ~ 17.09 and 0.262 5 ~ 16.80 respectively , 0.221 4 ~ 14.17,0.948 7 ~ 60.71,0.050 4 ~ 3.229,0.044 9 ~ 2.874 mg · L -1, the average recoveries were 100.4%, 99.8%, 100.2%, 100.0%, 99.0%, 98.9%, 97.4 % And RSD were 1.8%, 1.2%, 2.1%, 2.1%, 2.0%, 2.0% and 2.2% respectively. Conclusion: The method is simple and accurate. The contents of berberine, jatrorrhizine hydrochloride, berberine hydrochloride, palmatine hydrochloride, berberine hydrochloride, evodiamine and rutaecarpine in ethanol extract of Zujin pill are Zuo Jin Wan modern preparation research and quality control methodological reference.
