目的:研究雷丸多糖的提取、分离技术及其抗氧化活性。方法:用纤维素酶法酶解后对雷丸多糖进行提取工艺研究。通过Sevage法除蛋白,乙醇沉淀,活性炭脱色,再经Sephadex G-200柱层析法对其进行纯化,得到雷丸多糖OL1、OL2和OL3,并对其进行了抗氧化活性的研究。结果表明:酶解雷丸多糖的最佳提取工艺条件是:酶解温度40℃,酶添加量0.08%,酶解时间90min。结论:纤维素酶法可大大缩短雷丸多糖的提取时间,是极具应用前景的提取方法。雷丸多糖具有较强的清除自由基能力,是天然抗氧化剂的良好来源,可以进一步开发利用。
OBJECTIVE: To study the extraction and isolation technology of polysaccharide from Raymaru and its antioxidative activity. Methods: The extraction process of Leiwan polysaccharide was studied by enzymatic hydrolysis with cellulase. Proteins were removed by Sevage method, precipitated with ethanol and decolorized by activated charcoal. The polysaccharides were purified by Sephadex G-200 column chromatography to obtain OL1, OL2 and OL3 of Ray Broth, and their antioxidant activities were studied. The results showed that the optimal extraction conditions of polysaccharide extracted from Rayleigh pill were: enzymolysis temperature 40 ℃, enzyme dosage 0.08%, enzymolysis time 90min. Conclusion: Cellulase can greatly shorten the extraction time of Raymaru polysaccharide, and it is a promising method for extraction. Lei pill polysaccharide has a strong ability to scavenge free radicals, is a good source of natural antioxidants, which can be further exploited.
