目的 :研究大鼠局灶性脑缺血不同缺血时间皮质半暗带和中心区淀粉样前体蛋白 (APP)在转录水平表达规律。方法 :用线栓法建立大鼠局灶性脑缺血模型 ,剥取缺血半暗带及中心区皮质组织 ,采用半定量逆转录 聚合酶链式反应 (RT PCR) ,测定APPmRNA水平的变化。结果 :半暗带APPmRNA在缺血后 48h升高 ,缺血 72h达到高峰 ,缺血 1周后仍高于正常。缺血中心区APPmRNA在缺血后 72h和 96h高于正常水平。结论 :APPmRNA在缺血半暗带的表达上调 ,有可能加重缺血损伤。
Objective: To investigate the transcriptional expression of amyloid precursor protein (APP) in the cortex penumbra and central area of rats with focal cerebral ischemia and ischemia. Methods: The model of focal cerebral ischemia in rats was established by thread occlusion. The ischemic penumbra and central cortex tissues were stripped. The changes of APP mRNA level were determined by semi-quantitative reverse transcriptase-polymerase chain reaction (RT PCR) . RESULTS: APP mRNA in penumbra increased at 48h after ischemia and peaked at 72h after ischemia, still higher than normal one week after ischemia. APPmRNA in ischemic center was higher than normal at 72h and 96h after ischemia. Conclusion: APPmRNA is upregulated in ischemic penumbra, which may aggravate ischemic injury.
