目的研究淫羊藿苷(ICA)对阿尔茨海默病细胞模型中CREB表达的影响及其作用机制。方法以Aβ25-35孵育PC12细胞制备的AD细胞模型为研究对象,分为5组,分别为对照组、Aβ组、ICA、LY组、ICA+LY组(LY为PI3K/Akt信号通路的经典抑制剂),Western-blot法测定各组p-CREB(Ser133)/CREB、p-Akt(Ser473)/Akt的蛋白表达水平。结果 Aβ组中P-CREB、P-Akt蛋白表达水平明显低于对照组(P<0.01),且ICA组中P-CREB、P-Akt蛋白表达水平明显高于Aβ组(P<0.01),ICA+LY组中P-CREB、P-Akt蛋白的表达水平明显低于ICA组(P<0.01)。结论 Aβ25-35能明显下调PC12细胞中P-CREB、P-Akt蛋白表达水平,而ICA干预后能上调上述蛋白的表达水平。ICA可能通过PI3K/Akt信号通路来上调P-CREB表达水平而发挥其保护作用。
Objective To investigate the effect of icariin (ICA) on CREB expression in Alzheimer’s disease cell model and its mechanism. Methods AD cells cultured in PC12 cells incubated with Aβ25-35 were divided into 5 groups: control group, Aβ group, ICA, LY group and ICA + LY group (LY was the classical inhibition of PI3K / Akt signaling pathway) The protein expression levels of p-CREB (Ser133) / CREB and p-Akt (Ser473) / Akt in each group were determined by Western-blot. Results The expression of P-CREB and P-Akt in Aβ group was significantly lower than that in control group (P <0.01). The expression of P-CREB and P-Akt in ICA group was significantly higher than that in Aβ group The expression of P-CREB and P-Akt in ICA + LY group was significantly lower than that in ICA group (P <0.01). Conclusion Aβ25-35 can down-regulate the expression of P-CREB and P-Akt protein in PC12 cells, while ICA up-regulates the expression of these proteins. ICA may exert its protective effect by up-regulating the expression of P-CREB through the PI3K / Akt signaling pathway.
