目的观察胶质瘤中缺氧诱导园子-1α(HIF-1α)蛋白及HIF-1αmRNA的表达情况,探讨HIF-1α在胶质瘤中的表达特点,与胶质瘤分级的关系及意义。方法用SABC免疫组化法和原位杂交检测41例胶质瘤和8例正常脑组织标本中HIF-1α蛋白和HIF-1αmRNA的表达,并将胶质瘤分级,比较各级之间阳性表达率的差异。结果41例胶质瘤标本中HIF-1α蛋白阳性表达率为56.1%(23/41例),而8例正常恼组织中HIF-1α蛋白阳性表达率为0,二者相比有显著性差异(P<0.05)。HIF-1α蛋白在胶质瘤各病理分级中的阳性表达率分别为Ⅰ+Ⅱ级42.3%(11/26例),Ⅲ+Ⅳ级80.0%(12/15例),二者之间有显著性差异(P<0.05)。HIF-1αmRNA在胶质瘤中与正常脑组织表达无显著性差异(P>0.05),在胶质瘤各病理分级中的表达亦无显著性差异(P>0.05)。结论HIF-1α在胶质瘤中的表达上调,且与胶质瘤病理分级相关,它有助于肿瘤细胞在缺氧状态下的存活,协助肿瘤的生长和侵袭。HIF-1α在胶质瘤中表达的调节是在转录后水平。
Objective To observe the expression of HIF-1α and HIF-1αmRNA in gliomas induced by hypoxia, and to explore the relationship between the expression of HIF-1α in gliomas and the grade of glioma. Methods The expression of HIF-1α protein and HIF-1α mRNA in 41 gliomas and 8 normal brain tissues were detected by SABC immunohistochemistry and in situ hybridization. Gliomas were graded and the positive expression was compared among different grades Rate difference. Results The positive rate of HIF-1α protein expression in 41 glioma specimens was 56.1% (23/41), while the positive expression rate of HIF-1α protein in 8 specimens of normal angioma tissues was 0. There was a significant difference between the two (P <0.05). The positive rates of HIF-1α protein in glioma were 42.3% (11/26) and 80.0% (12/15) in grade Ⅰ + Ⅱ, respectively, there was significant difference between the two Sex differences (P <0.05). There was no significant difference in the expression of HIF-1αmRNA between normal gliomas and normal brain tissues (P> 0.05). Conclusions The expression of HIF-1α in glioma is up-regulated, and is related to the pathological grade of glioma. HIF-1α contributes to the survival of tumor cells under hypoxia and assists tumor growth and invasion. Regulation of HIF-1α expression in gliomas is at post-transcriptional level.
