目的:探讨大孔树脂纯化常春藤总皂苷的工艺条件。方法:选用AB-8和D101两种型号大孔吸附树脂,采用测定静态吸附量方法筛选树脂,并对总皂苷进行纯化,利用分光光度法测定纯化前后常春藤总皂苷的含量。结果:实验表明AB-8树脂的分离效果最好,其最佳工艺为上柱液流速3BV/h,样液浓度为2.00mg/mL,30%乙醇为洗脱液,洗脱液流速控制在2BV/h。在此条件纯化后,常春藤提取物中总皂苷含量由2%提高到37%。结论:AB-8大孔树脂可以较好地分离纯化常春藤总皂苷。
Objective: To investigate the process conditions of purifying total ivy saponins from macroporous resin. Methods: Two types of macroporous resin AB-8 and D101 were selected. The resin was screened by static adsorption capacity and the total saponin was purified. The content of total saponin before and after purification was determined by spectrophotometry. Results: The experiment results showed that AB-8 resin had the best separation efficiency. The optimum conditions were as follows: the flow rate of the column was 3BV / h, the concentration of the sample solution was 2.00mg / mL, the eluent was 30% ethanol, 2BV / h. After this condition was purified, the content of total saponin in ivy extract increased from 2% to 37%. Conclusion: AB-8 macroporous resin can be better purified ivy total saponins.
