目的研究重组人血清白蛋白-粒细胞集落刺激因子融合蛋白(GW003)对造血细胞的动员作用。方法 7只健康成年恒河猴单次皮下注射GW003 150μg/kg,分别于给药前后不同时间检测外周血细胞数和进行造血细胞集落培养。结果 GW003对健康成年恒河猴造血祖细胞、白细胞、中性粒细胞、未成熟粒细胞及单核细胞均有明显的动员作用,给药后18 h呈现高峰。注射GW003后10 h,外周血粒-巨噬系细胞集落形成单位(CFU-GM)、混合系细胞集落形成单位(CFU-Mix)、巨核系细胞集落形成单位(CFU-MK)数分别增至药前的1.8、1.9、1.4倍,红系爆式形成单位(BFU-E)和红系集落形成单位(CFU-E)数则增至1.7和1.5倍。结论 GW003对正常猴外周血干/祖细胞(PBS/PC)有明显动员作用,有望成为一种新的、高效的造血细胞动员剂。
Objective To study the mobilization of hematopoietic cells by recombinant human serum albumin-granulocyte colony-stimulating factor fusion protein (GW003). Methods Seven healthy adult rhesus monkeys were injected subcutaneously with 150 μg / kg of GW003, and the number of peripheral blood cells and the hematopoietic cell colony culture were detected at different times before and after administration. Results GW003 had a significant mobilization effect on hematopoietic progenitor cells, leucocytes, neutrophils, immature granulocytes and monocytes in healthy adult rhesus monkeys, and peaked at 18 h after administration. At 10 h after GW003 injection, the number of CFU-GM, CFU-Mix and CFU-MK in peripheral blood mononuclear cells increased to 1.8,1.9 and 1.4 times as much as pre-drug, the numbers of BFU-E and CFU-E increased to 1.7 and 1.5 times respectively. Conclusion GW003 has a significant mobilization effect on normal monkey peripheral blood stem / progenitor cells (PBS / PC) and is expected to become a new and effective hematopoietic mobilizing agent.
