纯钛超声工作尖与传统超声工作尖对钛表面形态及人牙周膜细胞黏附强度影响的比较研究

目的比较经纯钛超声工作尖与传统不锈钢超声工作尖刮治后的钛表面形态变化及人牙周膜细胞(human periodontal ligament cells,hPDLCs)对不同钛片表面的黏附强度。方法将60个实验所用钛片随机分为5组,每组12个。Ti10组:用纯钛超声工作尖刮治钛片表面10s;Ti30组:用纯钛超声工作尖刮治钛片表面30s;Cv10组:用传统超声工作尖刮治钛片表面10s;Cv30组:用传统超声工作尖刮治钛片表面30s;C组:钛片表面不做任何处理,作为对照。刮治后使用扫描电镜(scanning electron microscope,SEM)观察各组钛片表面的形态变化,再用表面粗糙度轮廓仪测量钛片表面粗糙度;将hPDLCs与处理后的各组钛片共同培养12h后,利用CCK-8法检测处理后钛片有无细胞毒性及hPDLCs的黏附强度。结果 SEM和表面粗糙度轮廓仪显示,经过表面处理后,Ti10组钛片表面粗糙度明显小于Cv10组(P<0.05),但增加处理时间至30s后各组之间的粗糙度差别无统计学意义(P>0.05)。细胞毒性实验表明,处理后的钛片无细胞毒性。处理后钛片的细胞黏附实验表明,hPDLCs对两种材料工作尖处理后的钛表面的黏附强度差异无统计学意义(P>0.05)。结论纯钛超声工作尖相比传统超声工作尖,在刮治过程中对钛表面的保护具有一定优势,但对刮治后的细胞黏附的促进,未显示出明显的优势。 Objective To compare the surface morphological changes of titanium treated with pure titanium ultrasonic tips and conventional stainless steel ultrasonic tips and the adhesive strength of human periodontal ligament cells (hPDLCs) on different titanium surfaces. Methods 60 experimental titanium tablets were randomly divided into 5 groups of 12. Ti10 group: using titanium ultrasonic tip scraping the surface of the titanium sheet 10s; Ti30 group: using pure titanium ultrasonic tip scrape titanium surface treatment 30s; Cv10 group: the use of conventional ultrasonic scraping scratch titanium surface 10s; Cv30 group: Using conventional ultrasonic tip scrape titanium surface treatment 30s; C group: titanium surface without any treatment, as a control. After the curettage, the morphological changes of the surface of each group were observed by scanning electron microscope (SEM), and then the surface roughness of the titanium was measured by surface roughness profilometer. The hPDLCs were incubated with the treated groups of titanium for 12h After using the method of CCK-8 detection of titanium after treatment with or without cytotoxicity and hPDLCs adhesion strength. Results SEM and surface roughness profiler showed that the surface roughness of Ti10 Ti alloy was significantly less than that of Cv10 (P <0.05) after surface treatment, but there was no significant difference between the groups after 30s increase of treatment time Significance (P> 0.05). Cytotoxicity experiments show that the treated titanium plate is non-cytotoxic. The results of cell adhesion experiments showed that the adhesion strength of hPDLCs to the titanium surfaces after two kinds of working tips had no significant difference (P> 0.05). Conclusion Titanium ultrasonic tip has some advantages over the traditional ultrasonic tip in protecting the titanium surface during the scraping and cure process. However, the promotion of cell adhesion after scraping does not show obvious advantages.
