目的: 了解肝纤维化大鼠肝组织Na+ /Ca2+泵(Na+ /Ca2+ exchanger,NCX)mRNA表达状况,探讨其与肝纤维化程度的关系及意义.方法: CCl4 sc法诱导大鼠肝纤维化,观察肝组织NCX及α1(Ⅰ)mRNA表达水平,检测血清HA,CⅣ,PCⅢ,血清肝纤维化相关酶(βNAG,MAO,ALD)、肝组织羟脯氨酸(hydroxyproline,Hyp)含量及肝组织病理学变化,分析其相关性与临床意义.结果: 20只正常大鼠肝组织NCXmRNA表达除1只弱阳性外,其余均为阴性,半定量积分仅为(0.10±0. 00);而30只模型组大鼠全部呈阳性,半定量积分显著增加(P< 0 .05 ).与正常组比较,模型组大鼠血清HA[ (455. 79±113 .55)vs(112 .41±45. 62)μg/L],CⅣ, [ (159. 62±29 64)vs(35 69±9 68)μg/L],PCⅢ[ (265 .54±98. 21)vs(89 .99±10. 85 ) μg/L]明显增高(均为P<0 01 );βNAG,MAO,ALD活性显著增加(分别增加2. 20, 2. 14, 1 .57倍,P<0 01),模型组大鼠NCXmRNA表达与α1(Ⅰ)mRNA表达水平、肝组织Hyp含量、血清HA,CⅣ,PCⅢ水平、肝组织纤维化积分均呈正相关,相关系数r分别为+0. 48, +0. 64, +0 .52,+0 49, +0 .56, +0 .60(均为P<0 05).结论: 肝纤维化大鼠肝组织NCXmRNA表达明显增强,与胶原代谢水平及肝脏组织病理学变化呈正相关,有望作为判断肝纤维化的有用指标,值得进一步?
Objective: To investigate the expression of Na + / Ca2 + exchanger (NCX) mRNA in hepatic tissue of rats with hepatic fibrosis and its relationship with the degree of hepatic fibrosis.Methods: CCl4 sc method was used to induce hepatic fibrosis in rats, The expression of NCX and α1 (Ⅰ) mRNA in liver tissues were observed. The levels of HA, CⅣ, PCⅢ, βNAG, MAO, ALD and hydroxyproline (Hyp) Pathological changes, analysis of their relevance and clinical significance.Results: NCXmRNA expression in 20 normal liver tissue except for a weak positive, the rest were negative, semi-quantitative integral was only (0.10 ± 0.00); 30 Only the model group rats were all positive, the semi-quantitative integral significantly increased (P <0 .05) .Compared with the normal group, serum HA [(455.79 ± 113.55) vs (112.41 ± 45 ), PCⅢ [(265 .54 ± 98.21) vs (89.999 ± 10.62) μg / L, CⅣ, [(15.69 ± 29.64) vs (P <0.01). The activities of βNAG, MAO and ALD increased significantly (by 2.20, 2.14 and 1.57 times, P <0.01) The expression of NCXmRNA and the expression of α1 (Ⅰ) mRNA, Hyp content in liver tissue, serum HA, CⅣ, PCⅢ The levels of fibrosis in liver and liver were positively correlated with the correlation coefficient r of +0.48, +0.64, +0.52, +0.49, +0.56, +0.60 (all P <0 05) .Conclusion: The expression of NCXmRNA in hepatic tissue of hepatic fibrosis rats is significantly increased, which is positively correlated with collagen metabolism and histopathological changes of liver, which is expected to be a useful index to judge hepatic fibrosis.
