目的 研究N Methyl D Aspartate(NMDA)受体拮抗剂MK 80 1对大鼠神经干细胞 (NSC)内源性激活的作用。方法 将不同年龄阶段 (10d、3月、10月 )的SD大鼠分为实验组和对照组 ,实验组腹腔注射MK 80 1,对照组腹腔注射生理盐水 ,用免疫组织化学方法测定两组大鼠海马齿状回颗粒层 (SGZ)的Brdu阳性细胞、Nestin阳性细胞表达数。结果 2组的 10d幼鼠 ,Brdu阳性细胞、Nestin阳性细胞增殖均不明显 ,2组无显著差异 (P >0 .0 5 ) ,而在 3月成年鼠实验组Brdu阳性细胞 7d达高峰 ,Nestin阳性细胞 11d达高峰 ,较对照组增殖较明显 (P <0 .0 5 ) ;实验组 10月老龄鼠Brdu阳性细胞、Nestin阳性细胞表达更明显 ,并可持续到 18d ,2组比较统计学有显著意义 (P <0 .0 1)。结论 NMDA受体拮抗剂MK 80 1可明显促进 3月成鼠、10月老年鼠脑中NSC的增殖、分化。
Objective To investigate the effect of N-methyl-D Aspartate (NMDA) receptor antagonist MK 80 1 on endogenous activation of rat neural stem cells (NSCs). Methods SD rats of different ages (10 days, 3 months, and 10 months) were divided into experimental group and control group. The experimental group was intraperitoneally injected with MK 80 1, the control group received intraperitoneal injection of normal saline and the immunohistochemical method was used to determine the difference between the two groups The numbers of Brdu positive cells and Nestin positive cells in SGZ of rat hippocampus. Results The proliferation of BrdU positive cells and Nestin positive cells were not significant in the two groups (P> 0.05), but no significant difference was found between the two groups (P> 0.05) (P <0.05). The positive expression of Brdu positive cells and Nestin positive cells in the experimental group were more obvious on the 11th day, which lasted for 18 days. The statistics of the two groups were statistically significant Significant significance (P <0. 01). Conclusion NMDA receptor antagonist MK 80 1 can significantly promote the proliferation and differentiation of NSC in the brain of adult rats in March and October.
