目的探讨银杏二萜内酯葡胺注射液对缺血/再灌注大鼠脑内氨基酸类和单胺类神经递质的影响。方法采用线栓法制备大鼠脑缺血/再灌注模型,缺血1.5 h,再灌注24h,尾静脉给予不同剂量银杏二萜内酯葡胺注射液后,用高效液相色谱串联电化学检测器(HPLC-ECD)测定脑内氨基酸类和单胺类神经递质含量。结果银杏二萜内酯葡胺注射液可以抑制缺血/再灌注导致的脑内天冬氨酸、谷氨酸、甘氨酸和γ-氨基丁酸的升高;降低缺血脑区皮层和海马内去甲肾上腺素、二羟苯乙酸、5-羟色胺、5-羟吲哚乙酸的浓度,抑制皮层内肾上腺素的减少。结论银杏二萜内酯葡胺注射液可以通过调节脑内氨基酸类和单胺类神经递质的水平,对大鼠脑缺血/再灌注引起的损伤发挥治疗作用。
Objective To investigate the effects of ginkgo diterpene lactone meglumine injection on amino acids and monoamine neurotransmitters in brain of ischemia / reperfusion rats. Methods The rat model of cerebral ischemia-reperfusion was established by thread occlusion. The rats were subjected to ischemia 1.5 h and reperfusion 24 h. After tail vein dosing with different dosages of diterpene lactam meglumine, the compounds were determined by high performance liquid chromatography tandem electrochemical (HPLC-ECD) for the determination of amino acids and monoamine neurotransmitters in the brain. Results Ginkgolide diterpene lactone meglumine injection could inhibit the increase of aspartate, glutamate, glycine and γ-aminobutyric acid in the brain caused by ischemia / reperfusion; reduce the cortex and hippocampus Norepinephrine, dihydroxybenzene acetic acid, serotonin, 5 - hydroxyindole acetic acid concentration, inhibit the reduction of cortical epinephrine. Conclusion Ginkgolic diterpene lactone meglumine injection can play a therapeutic role in the damage caused by cerebral ischemia / reperfusion in rats by regulating the levels of amino acids and monoamine neurotransmitters in the brain.
